A method of manufacturing a flexible container (1) housing a drug substance, comprises: forming a first compartment (11) of the container (1) out of a flexible sheet-like material, filling a liquid (2) into the first compartment (11) of the container (1), sealing the first compartment (11), forming a second compartment (12) of the container (1) out of the flexible sheet-like material, filling a dry drug formulation (3) into the second compartment (12), and sealing the second compartment (12). The method further involves lyophilizing the drug formulation inside a tubular cartridge such that the dry drug formulation (3) is generated and held in the tubular cartridge, wherein filling the dry drug formulation (3) into the second compartment (12) comprises: introducing the tubular cartridge holding the dry drug formulation (3) through an opening of the second compartment (12) of the container (1) such that an open end of the tubular cartridge is positioned distant from the opening of the second compartment (12), providing the dry drug formulation (3) out of the open end of the tubular cartridge into the second compartment (12), and withdrawing the tubular cartridge out of the opening of the second compartment (12) of the container (1). The first compartment (11) is separated from the second compartment (12) by a frangible seal (14) which opens when the first compartment (11) is compressed.

In this regard, the production system (10) uses a first conveyor (12) to sequentially load unit dose containers (130) into an empty container receptacle (24) of a second conveyor (20) at a first location (90). The second conveyor (20) may be operated to dispose a container receptacle (24) at each of the first location (90), a second location (100), and a third location (110). A unit dose container (130) at the second location (100) is evacuated and thereafter loaded with a radioactive fluid, and is thereafter moved to a third location (110) by the second conveyor (20). The unit dose container (130) is removed from the second conveyor (20) at the third location (110), and thereafter the radioactivity content of a unit dose container (130) may be determined, such as at an ionization chamber or the like.

These methods and systems can displace the pharmaceutical formulation through a density flow meter, wherein the density flow meter is configured to measure a density of the pharmaceutical formulation. Next, the pharmaceutical formulation can be dosed into preformed molds and the dosing process can be stopped when the density of the pharmaceutical formulation measured by the density flow meter is below a predetermined threshold.

Method, device and system for filling pharmaceutical containers

The present invention involves both an apparatus and an associated method for confirming the presence or absence of closures in a closure nest inside an aseptic chamber. The apparatus comprises a topographical profiler disposed to monitor the closure nests. Vertical displacements in topographical maps obtained by the profiler are compared with information from a database about the nest and closure combinations. Vertical displacements falling outside predetermined upper and lower bounds are deemed absences of closures. The topographical profiler may be disposed outside the aseptic chamber. The system may be automated via a controller and suitable software. In some embodiments the nests and disposed in the monitored area by robotic articulated arms. In other embodiments the closure nests are moved by vacuum pickup systems. The system and method may also be employed to assess the suitability of a closure nest for closing in an aseptic chamber containers in a container nest.
