Method for producing polyhydroxyalkanoates (PHA) from organic waste

According to one broad aspect of this disclosure, a method is provided for producing polyhydroxyalkanoates (PHA) from organic waste. The method comprises homogenizing organic waste to obtain a feedstock that has 1:1 to 3:1 (w/w) water to organic waste ratio. The feedstock is inoculated with an inoculum of acidogenic fermentative bacteria in order to obtain an inoculated feedstock. The inoculated feedstock is incubated for 5 to 10 days, 3 to 10 days, optionally 7 days, optionally 3 days, to obtain a fermentation broth. The fermentation broth comprises volatile fatty acids (VFAs) and undigested organic waste. The fermentation broth is filtered with a filter with a pore size ranging from 0.2 μm to 500,000 NMWC to remove the acidogenic fermentative bacteria and undigested organic waste, to obtain a clarified broth comprising concentrated VFAs. The clarified broth and high-PHA producing bacteria are incubated to produce intracellular PHA granules in the high-PHA producing bacteria. PHA polymers are extracted from the intracellular PHA granules.

A system and method of fluid delivery for providing a surface fluid pattern. The system includes a fluid delivery head for fluid flow therethrough. The fluid delivery head includes a fluid delivery surface having surface openings defined therein and arranged across the fluid delivery surface as a two-dimensional display. Some of the surface openings are grouped as a surface opening unit. The surface opening unit includes at least one aspiration opening through which fluid can be provided to the fluid delivery surface and at least one injection opening through which fluid can be moved away from the fluid delivery surface. The surface opening unit includes at least three surface openings positioned as a two-dimensional display and outwardly of at least one other surface opening.

To avoid liquid droplets or cell material being transported from one storage chamber to another as a result of a horizontal gas flow above the carrier during the treatment, the nozzles are arranged outside of and along an edge of the bearing region or within the bearing region and form a gas curtain around the carrier or the respective storage chambers.

A process and system for producing an inoculum for downstream cell production is disclosed. The inoculum is produced in a perfusion bioreactor in which the nutrient media feed is increased as the biomass concentration increases within the bioreactor. A biomass sensor can be used to periodically or continuously monitor biomass concentration. This information can be fed to a controller for automatically increasing nutrient media feed rates in a manner that is directly proportional to producing an inoculum with an increase cell density. The process and system can also include an automated subsystem for maintaining constant volume levels within the perfusion bioreactor during the process.
