MICROINJECTION CHIP, DEVICE, SYSTEM AND USES THEREOF FOR UNICELLULAR OR MULTICELLULAR ORGANISMS
Disclosed herein are microinjection chips, devices, and systems for injection of unicellular or multicellular organisms. The microinjection chip and device disclosed herein include the microfluidic features, inlet port, pre-injection reservoir, injection channel and post injection channel in fluid communication with each other. The inlet port is adapted to sequentially move individual organisms into the injection channel, which is adapted to immobilize the individual organism in fluid. The injection channel features a side wall adapted to receive a microinjection pipette without a microinjection port and to reseal when the microinjection pipette is removed.
Nanopipette apparatus for manipulating cells
Disclosed herein are methods and systems for controlled ejection of desired material onto surfaces including in single cells using nanopipettes, as well as ejection onto and into cells. Some embodiments are directed to a method and system comprising nanopipettes combined with an xyz controller for depositing a user defined pattern on an arbitrary substrate for the purpose of controlled cell adhesion and growth. Alternate embodiments are directed to a method and system comprising nanopipettes combined with an xyz controller and electronic control of a voltage differential in a bore of the nanopipette electroosmotically injecting material into a cell in a high-throughput manner and with minimal damage to the cell. Yet other embodiments are directed to method and system comprising functionalized nanopipettes combined with scanning ion conductance microscopy for studying molecular interactions and detection of biomolecules inside a single living cell.

An assembly and method to precisely and concurrently control the concentration, release, and depletion of chemical and biochemical species in localized regions in a three-dimensional volume of material using spatially patterned light. The system includes a container for holding a material comprising photo-responsive substance; the container is mounted to a motorized rotation stage that rotates the container along the z-axis at a predetermined rate; and a projector configured to project radiations at predefined wavelengths along an r-axis into the container while the container is rotating, wherein the radiations from multiple angles intersect to form localized photoreactive regions within a volume of the material.

An injector configured to inject a solution containing a biomolecule and a gas that is predetermined into an injection target without using an injection needle, the injector including a storage portion configured to store the solution containing the biomolecule and the gas, a nozzle portion communicating with the storage portion, the nozzle portion including an ejection port configured to eject the solution containing the biomolecule and the gas toward the injection target, and a pressurization portion configured to pressurize the solution containing the biomolecule and the gas that are stored in the storage portion during an operation and to eject the solution containing the biomolecule and the gas from the ejection port toward the injection target.

In various embodiments, method and devices for delivering large cargos (e.g., organelles, chromosomes, bacteria, and the like) into cells are provided. In certain embodiments method of delivering a large cargo into eukaryotic cells, are provided that involve providing eukaryotic cells disposed on one side of a porous membrane; providing the cargo to be delivered in a solution disposed in a reservoir chamber on the opposite side of the porous membrane; and applying pressure to the reservoir chamber sufficient to pass the cargo through pores comprising said porous membrane wherein said cargo passes through cell membranes and into the cells.
