Nucleic acid-guided nuclease editing in mammalian cells may include passaging mammalian cells, in an automated closed cell editing instrument, into smaller aggregates when the aggregates exceed 50-300 microns in size. A library of viral particles may be delivered to the mammalian cells at a multiplicity of infection such that each mammalian cell receives one or no viral particle. The library may include viral vectors with an editing cassette including a pair of gRNA coding sequence and donor DNA. Conditions may be provided to allow a viral vector of the viral vectors to integrate into the mammalian cells. Enriching for mammalian cells may be done with an integrated viral vector. A nucleic acid-guided nuclease or nuclease fusion or a coding sequence for a nucleic acid-guided nuclease or nuclease fusion may be delivered to the enriched mammalian cells and conditions may be provided to allow editing in the mammalian cells.

The stirring blade 14a is disposed between the thermal energy extracting pipes 12 and a rotating surface thereof is in a state of being parallel to the thermal energy extracting pipes 12. A bottom surface of the chip fermenter 11 is formed in an arcuate shape such as to follow the rotating outer peripheral surface of the stirring blade 14a. A plurality of temperature sensors 11a are installed in a vertical direction inside each chip fermenter. Control of an on-off valve of the thermal energy extracting pipe 12 is performed based on temperature information from the temperature sensors 11a.

The method also includes coupling electricity to the anode and the cathode, supplying carbon dioxide to the culture in the first chamber, and collecting methane from the culture at the outlet of the first chamber.

Embodiments of a system as discussed herein present an economically viable algae production system and process that allows algae-derived products such as fertilizer, feed, biofuels, etc. to compete with non-organic or petroleum products in the marketplace.
