COMPOSITIONS, SYSTEMS, AND METHODS FOR HIGH LEVEL EXPRESSION OF RECOMBINANT PROTEIN
Improved production of recombinant proteins in E. coli, reliant on tightly controlled autoinduction, triggered by phosphate depletion in stationary phase. The process also provides an optimized autoinduction media, enabling routine batch production at various culture volumes where cells densities routinely reach ˜5-7 g cell dry weight per liter and offer protein titers above 2 g/L. The methodology has been validated with a set of diverse heterologous proteins and is of general use for the facile optimization of routine protein expression from high throughput screens to fed-batch fermentation.
Method for producing animal model of preterm birth and animal model of preterm birth produced by the method
The present invention relates to a method for producing an animal model of preterm birth and an animal model of preterm birth produced by the method. The animal model of the present invention can be effectively applied to investigate the causes and symptoms of preterm birth induced by cervical injury. The mortality rate of the animal model according to the present invention is low until preterm birth despite its induced preterm birth. In addition, the animal model of the present invention is produced in a higher yield than any other existing model. Furthermore, the preterm birth of the animal model according to the present invention is induced at a desired time point. Due to these advantages, the animal model of the present invention can be effectively applied to investigate the causes and mechanisms of preterm birth. The mortality rate of premature neonates born from the animal model of the present invention is considerably low and the premature neonates are immature. Therefore, the animal model of the present invention can be effectively applied to studies on complications of premature neonates.
Microorganism strain for high-performance metabolism of biomass-derived carbon source
The present invention relates to a novel microorganism capable of metabolizing various carbon sources at high rates. A novel microorganism according to the present invention was observed to grow at a very high rate in a minimal medium/nutrient medium, etc., compared to microorganisms such as Escherichia coli, and shows resistance at a high initial sugar/salt concentrations as well as being able to produce lycopene and 2,3-butanediol through genetic manipulation. Therefore, the novel microorganism can be used in various production fields of high value-added compounds using microorganisms.
METHODS OF TREATING DEMENTIA ASSOCIATED WITH ALZHEIMER'S DISEASE WITH PROTECTIVE PROTEIN/CATHEPSIN A (PPCA)
Methods are provided for the prognosis, diagnosis and treatment of various pathological states, including cancer, chemotherapy resistance and dementia associated with Alzheimer's disease. The methods provided herein are based on the discovery that various proteins with a high level of sialylation are shown herein to be associated with disease states, such as, cancer, chemotherapy resistance and dementia associated with Alzheimer's disease. Such methods provide a lysosomal exocytosis activity profile comprising one or more values representing lysosomal exocytosis activity. Also provided herein, is the discovery that low lysosomal sialidase activity is associated with various pathological states. Thus, the methods also provide a lysosomal sialidase activity profile, comprising one or more values representing lysosomal sialidase activity. A lysosomal sialidase activity profile is one example of a lysosomal exocytosis activity profile.
COMPOSITE MATERIAL AND PREPARATION METHOD THEREFOR AND APPLICATION THEREOF
The present disclosure provides a composite material. The composite material comprises nanoparticles and a flexible substrate, the nanoparticles comprise one or more of carbon nanotubes, graphene, gold nanoparticles, and polydopamine nanoparticles, the flexible substrate comprises one or more of thermosetting plastics such as polydimethylsiloxane and a hydrogel, and the mass percentage of the nanoparticles in the composite material is 0 to 60‰. The composite material of the present disclosure is easy to prepare, has extremely strong photothermal conversion performance, and does not change the smooth surface of an original topological structure. Meanwhile, the composite material has universality and versatility for different cells, the delivery efficiency is close to 100%, and modified cells may be efficiently and non-destructively released and harvested by means of traditional trysinization, and the harvesting efficiency is 90% or more.
Nucleic acid construct
The present invention provides a nucleic acid construct comprising the following structure: A-X-B in which A and B are nucleic acid sequences encoding a first and a second polypeptide of interest (POI); and X is a nucleic acid sequence which encodes a cleavage site, wherein either the first or second POI is a transmembrane protein which comprises an intracellular retention signal.
SINGLE DOMAIN ANTIBODIES DIRECTED AGAINST INTRACELLULAR ANTIGENS
This invention provides compositions and methods to prevent aberrant cell proliferation in a subject using a single-domain antibody (sdAb) directed against an intracellular component, wherein the aberrant cell proliferation can be cancer. The sdAb is synergistic with one or more chemotherapeutic drugs and improves therapeutic efficacy of the one or more chemotherapeutic drug against cancer. The invention also includes a method of treating viral infections using a sdAb, wherein the sbAb inhibits the replication of viruses such as Ebola virus and Zika virus in infected cells.
