Method using a three-dimensional bioprocessor

Described herein is a beads-free bioprocessor as an automated and cost-effective T cell processing and manufacturing platform. T cells are a core component in CAR T cell therapies for cancer treatment, but are difficult to manufacture to scale in clinically relevant quantities. The 3D bioprocessor provides an alternative device that is scalable, beads-free, easy-to-use, and cost-effective for using CAR T cell therapy in cancer immunotherapy. Besides CAR T cell application, this platform technology has potential for many other applications such as cancer cell isolation.

Cortical spheroids and methods of making the same

Improved hybrid neurovascular spheroids and methods for making the same are provided. In some embodiments of a method for making a hybrid neurovascular spheroid, the method includes i) propagating cortical cells to form a cortical spheroid; ii) propagating endothelial cells to form an endothelial spheroid; iii) propagating mesenchymal stem cells to form a mesenchymal cell culture; and iv) combining the cortical spheroid, endothelial spheroid, and mesenchymal spheroid under conditions to form the hybrid neurovascular spheroid.

A method of efficiently recovering a large amount of exosomes from mesenchymal stem cells is provided. The method includes: a three dimensional culture step of three dimensionally culturing mesenchymal stem cells in a medium containing sugar by using a nonwoven fabric as a scaffold; a post-plateau culture step of further culturing for a certain period of time after the amount of the sugar consumed by the mesenchymal stem cells reaches a plateau; and an exosome recovery step of recovering exosomes from the mesenchymal stem cells. The mesenchymal stem cells are adipose-derived mesenchymal stem cells.

SYSTEM AND METHOD FOR ORGANOID CULTURE

The present disclosure provides a system, including methods and apparatus, for culturing, monitoring, and/or analyzing organoids. In an exemplary method of organoid culture, the method may comprise disposing a scaffold in a receptacle having an open side. A sealing member may be bonded to the open side of the receptacle to create a chamber. An organoid may be formed in the chamber using the scaffold. Fluid and/or at least one substance may be introduced into the chamber from an overlying reservoir for contact with the organoid.

The substrates, systems, and methods described herein relate to textured substrates for preparing a comestible meat product. Substrates and methods are described herein for controlling one or more of growth, adhesion, retention, and/or release of cells (e.g., of a cell sheet) on or from the surface of the substrate. A method of preparing a comestible meat product may include applying a plurality of non-human cells to at least one patterned texture substrate, growing the cells on the patterned texture substrate to form the comestible meat product, and separating the comestible meat product from the patterned texture substrate. The patterned texture allows for improved growth, adhesion, retention, and/or release of cells as compared to another surface not comprising the patterned texture. In some embodiments, the cell culture substrate surfaces include a plurality of regions corresponding to a plurality of patterned textures.
