Methods, cells and reagents for production of isoprene, derivatives and intermediates thereof
This application describes methods, including non-naturally occurring methods, for biosynthesizing 3-hydroxy-3-methylglutaryl-coA and intermediates thereof, as well as non-naturally occurring hosts for producing 3-hydroxy-3-methylglutaryl-coA. This application also describes methods, including non-naturally occurring methods, for biosynthesizing isoprene and intermediates thereof, as well as non-naturally occurring hosts for producing isoprene.
Neurotransmitters and Methods of Making the Same
In an aspect, the disclosure provides methods for making neurotransmitters in a host organism. The neurotransmitters can be cannabinoids and derivatives of cannabinoids. The host cells can be microalgae, fungi or other host cells. In a related aspect, the disclosure provides host cells engineered to have biochemical pathways for making neurotransmitters such as cannabinoids.
HIGH EFFICIENCY PRODUCTION OF CANNABIGEROLIC ACID AND CANNABIDIOLIC ACID
The present disclosure features compositions and methods for producing one or more cannabinoids, such as cannabidiolic acid (CBDa) or cannabidiol (CBD), in a host cell, such as a yeast cell, that is genetically modified to express the enzymes of a cannabinoid biosynthetic pathway. Using the compositions and methods of the present invention, the host cell may be genetically modified to express one or more enzymes of a cannabinoid biosynthetic pathway, such as CBDaS. The host cell may be cultured in the medium in the presence of a first cannabinoid, for example CBGa, and incubated for a time sufficient to allow for bioconversion of the first cannabinoid to a second cannabinoid, for example CBDa, by the host cell.
ORGANISMS PRODUCING LESS CROTONIC ACID
The present invention relates to a recombinant organism or microorganism having a decreased pool of crotonic acid compared to the organism or microorganism from which it is derived due to at least one of: (i) an increased conversion of crotonyl-CoA into butyryl-CoA; and/or an increased conversion of butyryl-CoA into butyric acid; (ii) an increased conversion of crotonyl-CoA into 3-hydroxybutyryl-CoA; and/or an increased conversion of 3-hydroxybutyryl-CoA into 3-hydroxybutyric acid; (iii) an increased conversion of crotonic acid into crotonyl-CoA; (iv) an increased conversion of crotonyl-[acyl-carrier protein] into butyryl [acyl-carrier-protein]; (v) a decreased conversion of crotonyl-CoA into crotonic acid; and/or (vi) a decreased conversion of crotonyl-[acyl-carrier protein] into crotonic acid. Moreover, the present invention relates to the use of such a recombinant organism or microorganism for the production of alkenes with the enzyme ferulic acid decarboxylase. Further, the present invention relates to a method for the production of isobutene or butadiene by culturing such a recombinant organism or microorganism in a suitable culture medium under suitable conditions.
