EXTRACELLULAR VESICLE-BASED NANOCARRIERS
Disclosed herein is a system that engages skin-resident APCs by directly delivering a vaccine composition, and a system that turns skin cells into a vaccine dispatch center to amplify immunity via the production of engineered extracellular vesicles (EVs) functionalized with targeting ligands and loaded with the vaccine composition that can be targeted to extracutaneous APCs. In particular, disclosed herein is a vaccine composition that involves a first polynucleotide encoding or comprising a viral, bacterial, or tumor antigen, and a second polynucleotide encoding a fusion protein comprising an APC-targeting ligand and an exosomal or lysosomal transmembrane protein. Also disclosed is a method of vaccinating a subject that involves transfecting skin cells of the subject with the disclosed vaccine composition. Also disclosed herein is a method of vaccinating a subject that involves administering to the subject the disclosed EV vaccine.

Provided herein are therapeutic agents comprising an elastin-like peptide (ELP) component and a ligand that specifically targets and binds an ICAM-1 receptor. In one aspect, the ELP comprises, or alternatively consists essentially of, or yet further consists of one or more sequence(s) designated S48I48 and/or mICMA-1 SI and/or hICAM-1 S1 or biological equivalents thereof. In a further aspect, the ELP/ligand composition further comprises a therapeutic agent. Methods and compositions for natural killer cells
Disclosed herein are novel compositions and methods for stimulation of and the production or expansion of natural killer (NK) cells. Numbers of NK cells can be increased following contact with exosomes modified with one or more stimulatory peptides. Methods and compositions for the production of exosomes, wherein the exosomes comprises stimulatory peptides are also described. Also described are methods of treating cancer using the disclosed NK-stimulating exosomes or NK cells stimulated by the disclosed methods.
