Methods, Systems, and Apparatus for Performing Flow Cytometry
An apparatus for identifying analytes of a first type in a fluid sample having a plurality of types includes a closed flow cell. The flow cell includes a sample inlet, a sample outlet. A flow path, between the inlet and outlet, passing through an interrogation region and at least a portion extending from immediately upstream to immediately downstream of the interrogation region is bounded by a flow cell well to form a continuous closed flow path. The apparatus also includes a spherical element having a center point coincident with the interrogation region. A hydrodynamic focusing element upstream of the interrogation region creates a core-in-sheath flow along a flow axis of the flow path as it passes through the interrogation region. Further, the apparatus includes a concave, ellipsoidal collector element having a first focal point disposed in the interrogation region of the flow path. A plurality of beams of electromagnetic energy converge non-axisymmetrically at the interrogation region, each of the plurality of beams orthogonal to the flow axis.
Use of Vibrational Spectroscopy for DNA Content Inspection
This disclosure concerns a cytometry system including a handling system that enables presentation of single cells to at least one laser source. The laser source is configured to deliver light to a cell within the cells in order to induce bond vibrations in the cellular DNA. The system further includes a detection facility that detects the signature of the bond vibrations, wherein the bond vibration signature is used to determine the folding or packing of the DNA.

By subjecting the sperm population to artificial insemination or in vitro fertilization, gender preselection for obtaining male offspring is possible.
Methods, systems and apparatus for performing flow cytometry
An apparatus for detecting an analyte in a sample includes an illumination source for generating electromagnetic energy to illuminate the sample at an interrogation region, a concave collector element having an optical axis, and a focal point, the interrogation region being coincident with the focal point of the concave collector element, a closed flow cell having a flow path defined between a sample inlet and a sample outlet, the flow path passing through the interrogation region and a sorting region disposed downstream of the interrogation region. The portion of the flow path passing through the interrogation region is coaxially aligned with the optical axis of the concave collector element. The apparatus further includes the sample comprising or suspected of comprising the analyte and flowing in the flow path, wherein the analyte generates a detectable signal in response to illumination. The apparatus also includes a detector for detecting the detectable signal.

Solution to Problem The present disclosure relates to a male transgenic non-human animal in which one of an X chromosome and a Y chromosome comprises an exogenous embryonic death induction gene operatively connected to a drug-responsive promoter. The present disclosure also includes a method for producing a male transgenic non-human animal for sex selection.

The present invention provides an aptamer or pool of aptamers (nucleic acid sequences) that bind(s) to a target molecule on the surface, accessible from the surface or inside of a mammalian sperm cell and a method for producing the aptamers. The method comprises contacting a collection of different nucleic acid molecules with the target molecule under conditions favorable for binding between at least one of the nucleic acid molecules and the target molecule, to form at least one complex comprising the nucleic acid molecule bound to the target molecule, wherein each of the nucleic acid molecules comprises at least one segment of randomized nucleotide sequences. The complexes are then separated from the unbound nucleic acid molecules and unbound target molecules, and the bound nucleic acid molecule is recovered from the separated complex. The aptamers are used to separate sperm cells based upon sperm cell qualities, such as whether the cells contain X chromosomes or Y chromosomes.
