Humanized mouse model
A non-human mammalian model for human diseases or disorders comprising a non-human neutrophil depleted mammalian host engrafted with a human skin equivalent (huSE) and human immune cells.
Dendritic Cell Generator
Devices, systems, and methods can be used for the automated production of dendritic cells (DC) from dendritic cell progenitors, such as monocytes obtained from peripheral blood. The invention makes it possible to obtain sufficient quantities of a subject's own DC for use in preparing and characterizing vaccines, for activating and characterizing the activation state of the subject's immune response, and to aid in preventing and/or treating cancer or infectious disease.
Engineered Artificial Antigen Presenting Cells for Tumor Infiltrating Lymphocyte Expansion
In some embodiments, compositions and methods re¬lating to isolated artificial antigen presenting cells (aAPCs) are dis¬closed, including aAPCs comprising a myeloid cell transduced with one or more viral vectors, such as a MOLM-14 or a EM-3 myeloid cell, wherein the myeloid cell endogenously expresses HLA-A/B/C, ICOS-L, and CD58, and wherein the one or more viral vectors com¬prise a nucleic acid encoding CD86 and a nucleic acid encoding 4-1BBL and/or OX40L and transduce the myeloid cell to express CD86 and 4-1BBL and/or OX40L proteins. In some embodiments, methods of expanding tumor infiltrating lymphocytes (TILs) with aAPCs and methods of treating cancers using TILs after expansion with aAPCs are also disclosed.
Lipids as synthetic vectors to enhance antigen processing and presentation ex-vivo in dendritic cell therapy
The invention covers the use of certain classes of lipids including cationic lipids in ex-vivo dendritic cell therapies. The cationic lipids enhance antigen uptake, processing and presentation of the processed antigens by dendritic cells to CD8+ and CD4+ T-cells via the MHC classes I and II presentation pathways respectively. Antigen uptake via cationic lipid by dendritic cells result in significant lowering of the population of the immune suppressive regulatory T cells in the tumors and a significant increase of the tumor targeting cytotoxic T-cells. Loss of regulatory T cells and increase of tumor specific cytotoxic cells are conducive to effective elimination of the tumors.
Method of producing immunotherapy vaccine
A method of producing an immunotherapy vaccine is provided. The method includes performing a heat treatment to exosomes separated from cancer cells or body fluids including blood of a cancer patient to promote inactivation of proteolytic enzymes in the exosomes, and introducing or co-culturing the exosomes in dendritic cells derived from blood of the cancer patient or a healthy person to make antigen-presenting cells.
DIFFERENTIATION OF PRIMATE PLURIPOTENT STEM CELLS TO HEMATOPOIETIC LINEAGE CELLS
The invention provides methods of differentiating primate pluripotent stem cells into cells of hematopoietic lineage. The invention further provides hematopoietic lineage cells differentiated from primate pluripotent stem cells, as well as methods of using the same and kits comprising the same.
Enhancing the T-cell stimulatory capacity of human antigen presenting cells in vitro and in vivo and its use in vaccination
We provide new methods of in vitro or in vivo enhancing the T-cell stimulatory capacity of human DCs and the use thereof in cancer vaccination. The method includes the introduction of different molecular adjuvants to human DCs by contacting or modifying them with mRNA or DNA molecule(s) encoding CD40L, and CD70 or constitutively active TI R4 (caTIR4).
Composition containing complex cytokines derived from EBV-infected B cells for inducing the maturation of dendritic cells
A composition for deriving the maturation of dendritic cells includes complex cytokines generated by the simulation, the expression of which is induced on EBV-infected B cells. The dendritic cell maturation process, which conventionally takes approximately 7 days, can be shortened to 2 days, thereby producing dendritic cells in a more economically advantageous and effective manner.
Method for generating human dendritic cells for immunotherapy
In various embodiments methods of producing a cell population enriched for CLEC9A+ dendritic cells are provided where the methods involve culturing stem cells and/or progenitor cells in a cell culture comprising culture medium, a notch ligand, stem cell factor (SCF), FLT3 ligand (FLT3L); thrombopoietin (TPO); and IL-3 and/or GMCSF.
Tolerogenic dendritic cells for treating myocardial infarction and manufacturing method thereof
Provided are the tolerogenic dendritic cells for treating a myocardial infarction and a method for preparing the same, and more particularly, the tolerogenic dendritic cells for treating a myocardial infarction, which can treat cardiac insufficiency that occurs by excessive remodeling of left ventricle in the heart muscle recovery process after an acute myocardial infarction, and a method for preparing the tolergenic dendritic cells. According to the present invention, the inflammatory reaction and the excessive remodeling of left ventricle in the heart muscle recovery process after a myocardial infarction can be inhibited, and thus, the incidence rate of cardiac insufficiency can be significantly reduced, thereby being effectively used for treating a myocardial infarction.
