RECOMBINANT ADENOVIRUSES AND USES THEREOF
Featured are recombinant adenoviruses and vectors thereof. In particular, the adenoviruses are simian (rhesus) adenoviruses having a low seroprevalence and high immunogenicity (when expressing, e.g., an antigenic polypeptide) relative to other adenoviruses and vectors thereof. Also featured are methods for producing the adenoviruses and methods of treatment of diseases by administering the adenoviral vector(s) to a subject (e.g., a human).
ONCOLYTIC VIRUSES AS ADJUVANTS
Herein is described oncolytic viruses for use as immunologic adjuvants. There is provided a method of adjuvanting an immune response to an antigenic protein in a mammalian subject by administering the oncolytic virus and at least one antigenic peptide, with the latter not encoded by the former. Without the requirement for the virus to encode the antigenic protein, therapies may be readily personalized or formulated. The virus may be attenuated or inactivated. Prime:boost therapies for tumours are also provided, in which the prime comprises at least one antigenic protein, the boost comprises a virus and at least one antigenic protein, the at least one antigenic protein of the prime and the at least one antigenic protein of the boost are based on the same at least one tumour associated antigen, and the at least one antigenic protein of the boost is not encoded by the virus of the boost.
METHOD TO PRODUCE A MEDICINAL PRODUCT COMPRISING A BIOLOGICALLY ACTIVE PROTEIN AND THE RESULTING PRODUCT
The present invention pertains to a method for producing a medicinal product comprising a biologically active protein comprising the steps of providing an aqueous composition comprising a solvent, the biologically active protein and between 20% w/w and 60% w/w of a non-polymeric sugar, freezing the composition, thereby forming at least one frozen body comprising the solvent in frozen form, putting the frozen body in a drying apparatus while being carried by a support, the support comprising one or more restraining elements that define one or more boundaries of the support, wherein at most 30% of the surface of the body is contiguous with the one or more restraining elements, reducing the pressure in the drying apparatus below atmospheric pressure, providing heat to the body in order to sublimate the frozen solvent of the body and obtain a dried body. The invention also pertains to a product obtainable by this method.
COMPOSITIONS AND METHODS FOR VIRAL SENSITIZATION
Provided are compounds that enhance the efficacy of viruses by increasing spread of the virus in cells, increasing the titer of virus in cells, or increasing the antigen expression from a virus, gene or trans-gene expression from a virus, or virus protein expression in cells. Other uses, compositions and methods of using same are also provided.
ADENOVIRAL COAT PROTEIN DERIVED DELIVERY VEHICLES
The present invention relates to new adenoviral coat protein based delivery vehicles. They are based on a modified penton base protomers that assemble into VLPs. Exposed areas of the penton base proteins can be modified to allow the VLP to specifically bind to any target and/or to comprise any desired peptide epitope. Additional cargo, e.g. drugs, proteins, or nucleic acids, can reversibly or irreversibly attached to the VLP via engineered fibre protein fragments. The present invention relates to such engineered penton base protomers, engineered proteins comprising an adenovirus fibre protein N-terminal fragment specifically binding to an adenovirus fibre protein binding cleft of a penton base protomer, VLPs comprising the engineered penton base protomers and optionally engineered proteins comprising an adenovirus fibre protein N-terminal fragment specifically binding to an adenovirus fibre protein binding cleft of a penton base protomer, nucleic encoding the engineered proteins, the VLPs as well as methods of producing the proteins and VLPs.
ADENOVIRUS AND USES THEREOF
Provided herein are adenoviral nucleic acid sequences and adenoviral vectors comprising said nucleic acid sequences. The provided adenoviral vectors can be used to induce a protective immune response in a subject.
Methods Of Vaccine Administration
This invention relates to a method of treating a dog for canine diseases comprising administering to the dog therapeutically effective amounts of a vaccine, wherein the vaccine comprises viral antigens, a bacterin, or both, and wherein the vaccine is administered subcutaneously or orally according to the schedules provided herein.
Affenadenovirus (gorilla) or adenoviral vectors and methods of use
The invention provides an adenovirus or adenoviral vector characterized by comprising one or more particular nucleic acid sequences or one or more particular amino acid sequences, or portions thereof, pertaining to, for example, an adenoviral pIX protein, DNA polymerase protein, penton protein, hexon protein, and/or fiber protein.
SEQUENCE ARRANGEMENTS AND SEQUENCES FOR NEOEPITOPE PRESENTATION
Systems and methods are presented that allow for selection of tumor neoepitopes that are then used to generate a recombinant polytope that is optimized for proper trafficking and processing. In preferred methods, the polytope is encoded in a viral expression system that is used as a therapeutic agent.
SIMIAN ADENOVIRAL VECTORS WITH TWO EXPRESSION CASSETTES
A simian adenoviral vector comprising two expression cassettes, wherein each expression cassette comprises a transgene and a promoter, and wherein the first expression cassette is inserted in the E1 region of the simian adenoviral vector, and the second expression cassette is inserted in a region of the adenoviral vector that is compatible with vector replication.
