Provided herein are engineered microorganisms comprising one or more genes involved in fatty acid metabolism. For example, provided is an engineered microorganism comprising a first nucleic acid sequence encoding an elongase and a second nucleic acid sequence encoding a desaturase, wherein the first and second nucleic acid sequences are operably linked to promoter. Also provided are methods of making and using the engineered microorganisms. Also provided are microbial oils comprising fatty acids, wherein the fatty acids comprise C20:3(n-6) (di-homo-?-linoleic acid) and C20:5(n-3) eicosapentaenoic acid (EPA). In addition, provided herein are methods of promoting conversion of saturated fatty acids to unsaturated fatty acids by transforming microorganisms with nucleic acid encoding polypeptides involved in the fatty acid synthesis pathway to generated increased conversion of saturated fatty acids to unsaturated fatty acids.

Provided herein are recombinant microorganisms having two or more copies of a nucleic acid sequence encoding xylose isomerase, wherein the nucleic acid encoding the xylose isomerase is an exogenous nucleic acid. Optionally, the recombinant microorganisms include at least one nucleic acid sequence encoding a xylulose kinase and/or at least one nucleic acid sequence encoding a xylose transporter. The provided recombinant microorganisms are capable of growing on xylose as a carbon source.

Linoleic acid isomerase and its application in production of conjugated linoleic acid
Disclosed is linoleic acid isomerases and their application in production of conjugated linoleic acid, which belongs to the technical fields of protein engineering and microbial engineering. The linoleic acid isomerase derived from Bifidobacterium is used to produce the conjugated linoleic acid. The recombinant E. coli containing the linoleic acid isomerase of the invention is added into a reaction system containing linoleic acid and react for 3 h to produce conjugated linoleic acids. The conversion rate of the conjugated linoleic acid of the invented method ranges from 12.1% to 42.1%, and the percentage of cis9, trans11-CLA in the conjugated linoleic acid can reach 84.3% to 89.1%. The invention provides a method for using microorganisms to produce conjugated linoleic acids with high safety and yield where cis9, trans11-CLA isomer is the major form in the conjugated linoleic acid products.

Provided herein are recombinant microorganisms having two or more copies of a nucleic acid sequence encoding xylose isomerase, wherein the nucleic acid encoding the xylose isomerase is an exogenous nucleic acid. Optionally, the recombinant microorganisms include at least one nucleic acid sequence encoding a xylulose kinase and/or at least one nucleic acid sequence encoding a xylose transporter. CONJUGATED LINOLEIC ACID-PRODUCING STRAINS OF PROBIOTIC BACTERIA AND USE THEREOF FOR THE PREPARATION OF A FOOD, DIETETIC OR PHARMACEUTICAL COMPOSITION
The present invention relates to conjugated linoleic acid (CLA)-producing strains of probiotic bacteria. In particular, the present invention relates to a selection of bacterial strains belonging to the genus Bifidobacterium which were selected for their ability to produce conjugated linoleic acid (CLA) from linoleic acid (LA). Furthermore, the present invention relates to a food, dietetic or pharmaceutical composition comprising said bacterial strains capable of increasing the quantity of CLA in situ, i.e. inside the gastrointestinal tract.
