Sustained release composition

The present invention relates to sustained release compositions, and in particular a sustained release composition comprising spermatozoa embedded in a matrix, such as a polymeric matrix. By having at least two sections with distinct dissolution rate, the matrix will ensure that spermatozoa are released for a prolonged period of time and thus making the time of insemination relative to ovulation less critical. Further, the present invention also relates to a method for preparing the sustained release composition, container comprising the sustained release composition and use of the sustained release composition in the breeding of animals.

An apparatus is provided for use with a fluid that contains biological cells, e.g. seminal fluid. The apparatus (20) includes a tray (22) shaped to define a flat upper surface configured to support the fluid, and an underside. The apparatus (20) further includes a plurality of protrusions protruding from the underside of the tray, which may serve to separate the underside from a flat surface on which the tray is placed, or as delineating protrusions that facilitate subsequent retrieval of the fluid following placement on the tray. The tray has a thickness between 0.4 and 0.8 mm, and is transparent or translucent. The upper surface preferably has a level of hydrophobicity that is sufficient for droplets to maintain their shape to a certain extent. A handle (52) facilitates handling of the tray. The apparatus typically fits into a standard-sized cryogenic vial (70).

A method of cryopreservation of biological material includes the steps of providing an apparatus including a vessel having a reservoir wherein the vessel has a channel formed in a portion of the reservoir, the channel including an intermediate restriction dimensioned to resist passage of the biological material but allow passage of liquid treatment solutions, and the channel including sub-reservoirs at either end of the restriction; introducing the biological material into a reservoir region of the apparatus for capture in a channel region of the vessel; irrigating and draining the biological material with a series of vitrification solutions introduced into a first of the sub-reservoirs and removed from a second of the sub-reservoirs; final draining the irrigation solutions from the vessel; and vitrifying the vessel and captive biological material.

For one or preferably for each of said sheets (11), said layer of weldable thermoplastic material (35) comprises triclosan; and the ratio between said predefined volume of said pouch and the sum of the areas of the surfaces (30) of said sheets (11) facing said pouch is between 0.3 ml/cm.sup.2 and 0.5 ml/cm.sup.2. The system comprises such bags and an antibiotic-free dilution medium.
