Oral delivery for hemoglobin based oxygen carriers

A process for making hemoglobin based oxygen carrier (HBOC) containing pharmaceutical composition suitable for oral delivery and the composition formed thereby are described. There are three exemplary composition configurations which include (1) hemoglobin-loaded nanoparticles solution, (2) enteric-coated hemoglobin capsules and (3) enteric-coated hemoglobin tablets. To facilitate the bioavailability and bio-compatibility of hemoglobin, intestinal absorption enhancers are added in each of the HBOC formulations. Protective layers ensure delivery of an intact hemoglobin structure in intestinal tract without degradation in the stomach. The HBOC formulations may be used for preventive or immediate treatment of high altitude syndrome (HAS) or for treatment of hypoxic conditions including blood loss, anemia, hypoxic cancerous tissue, and other oxygen-deprivation disorders. In addition to delivering oxygen, the heme group of hemoglobin from HBOC formulations can provide heme iron to the human body to aid in the production of more red blood cells.

Gels are formed based on generally recognized as safe (GRAS) low molecular weight amphiphilic molecules in a self-assembly process with limited or no heating. A selective range of ratios between an organic solvent and water, or an aqueous solution, in the medium, allows for the GRAS low molecular weight amphiphiles to form a homogeneous self-supporting gel encapsulating agents to be delivered under very mild conditions. Proteins including enzymes, antibodies, and serum albumin are loaded in the self-assembled gels to provide sustained and/or responsive delivery. The encapsulated proteins retain at least 70%, 80%, or 90% of their activity over days in various storage conditions.

A self-assembled gel composition with enhanced adhesion to cartilage tissue is provided. A cationic agent co-self assembles with a generally regarded as safe (GRAS), low molecular weight (<2,500 Da) gelator, forming homogeneous self-supporting gel that can encapsulate one or more therapeutic agents for controlled release. The composition adheres to connective tissue, e.g., cartilage, to a greater extent and a greater length of time than a self-assembled gel from gelators alone. The composition is used to specifically target connective tissue and deliver one or more therapeutic, prophylactic, or diagnostic agents for controlled release to improve dosing efficacy.

Process for Producing a Soft Gel Capsule Comprising Viable Probiotic Bacteria and a Soft Gel Capsule Comprising Viable Probiotic Bacteria Having a Long Shelf Life

The present invention provides a method for producing a soft gel capsule comprising uncoated probiotic bacteria, the method comprising gentle mixing the uncoated probiotic bacteria with at least one oil to obtain a soft gel capsule fill material at a temperature in the range of 5 to 15° C., encapsulating the fill material in a soft gel capsule made of a gelatin having a melting point in the range of 11 to 28° C.; and drying the soft gel capsule in one or more steps to a water activity of at the most 0.25 at a temperature of at the most 25° C., as well as soft gel capsules produced by this method. The present invention further provides a soft gel capsule comprising dried, uncoated, non-spore-forming probiotic bacteria wherein the soft gel capsule comprises at least E+09 viable bacteria after 24 months of storage at 25° C., e.g. at least 2 E+09 viable bacteria after 12 months of storage at 25° C.
