The glycosylated proteins may be synthesized in cell-free glycoprotein synthesis (CFGpS) systems using prokaryote cell lysates that are enriched in components for glycoprotein synthesis such as oligosaccharyltransferases (OSTs) and lipid-linked oligosaccharides (LLOs) including OSTs and LLOs associated with synthesis of bacterial O antigens.

Vaccine delivery method

A vaccine delivery method is presented that includes a composition including as one component a slurry matrix that is a liquid at room temperature and a gel at physiological pH, physiological salt concentrations and/or physiological temperatures and as a second component one or more antigens. Also included are methods of inducing an immune response in a subject and vaccinating a subject by administering such compositions.

A method for treating and/or mitigating a respiratory condition in an equine patient. The method comprises administering a blood product to the equine patient via localized delivery to the lungs. The blood product is produced by pre-treating at least one donor horse with a diet and immunization regime for at least one month before collecting the blood from the donor horse and processing it to make the blood product. The pre-treatment diet comprises feeding the donor horse a combined amount of EPA and DHA of at least 4.25 mg/kg horse/day with the EPA:DHA ratio ranging from 1:5-5:1 and alfalfa hay. The immunization regimen comprises immunizing the donor horse with Rhodooccus equi.

Ex vivo antigen and adjuvant pulsed peripheral blood mononuclear cells as a screening platform for candidate novel vaccines and candidate antigens

Peripheral blood mononuclear cells (PBMCs) can be used in place of DCs when pulsing with antigens, or antigen and adjuvant combination, and then administered to a subject as a vaccine to induce a protective immune response. The PBMC-based vaccine strategy provides a more marked and enduring protective immune response and is also capable of serving as a multi-organism prophylactic vaccine platform. The vaccine platform may be used to screen vaccine and adjuvant combinations and may also be used to allow for adjuvants that are otherwise unsafe for use in humans as the adjuvant may be removed prior to prophylactic administration of the pulsed PBMCs.

A system and method for prediction of immunodominant epitopes is provided herein. MHCII peptidomics was used to discover complex bacterial epitopes and host antigen processing pathways. Novel insights into the features of antigenicity are leveraged to build an algorithm for prediction of immunodominant epitopes. Use of immunodominant epitopes is described.
