Plasmid-based CTX phage replication system and vibrio cholerae strain that can be infected by CTX phage and can be used for cholera toxin production
The present invention relates to a plasmid-based CTX phage replication system and Vibrio cholerae strain that can be infected by CTX phage and can be used for cholera toxin production. More particularly, the present invention provides a Vibrio cholera variant strain, which expresses a toxT protein in which tyrosine at position 139 is substituted by phenylalanine through the point mutation of a toxT gene using a plasmid-based CTX phage replication system, and is used as a receptor strain which can improve CTX phage infection efficiency and allows a plurality of CTX prophages to simultaneously infect the strain and to be inserted into the chromosome thereof, which the consequent provision of the effect of increasing the production yield of a cholera toxin.
Bacterial polysaccharide-conjugated carrier proteins and use thereof
Immunogenic compositions that include a bacterial capsular polysaccharide conjugated to a carrier protein are described. In some embodiments, the bacterial capsular polysaccharide is a Neisseria meningitidis capsular polysaccharide. The carrier protein includes an N. meningitidis factor H binding protein (fHbp) linked to cholera toxin subunit B (CTB). Administration of the immunogenic compositions elicits an immune response that includes production of meningococcal polysaccharide-specific and fHbp-specific antibodies. Use of the immunogenic compositions as meningococcal vaccines is also described.
METHODS AND COMPOSITIONS RELATED TO THE NEXT GENERATION VACCINE
Disclosed are methods and compositions related to polypeptides comprising a fusion of the needle tip protein and translocator protein of a type III secretion apparatus (T3SA) from a type III secretion system (T3SS) of a Gram negative bacteria. Disclosed herein are fusion polypeptides comprising a fusion of a needle tip protein, such as, Bsp22, LcrV, BipD, PcrV, CT053, or CT668, or anantigenic fragment thereof; and a translocator protein, such as, BopB, YopB, BipB, PopB, CopB, or CopB2, or anantigenic fragment thereof from a Type III secretion system (T3SS) of a Gram negative bacteria, such as, Bordetella, Burkholderia, Chlamydia, Pseudomonas, Vibrio, or Yersinia.
IMMUNE-ENHANCED AQUACULTURE
Aquaculture systems and methods are provided, as well as immunogenic compositions and methods of immunologically protecting bivalves against specified pathogens. Methods include inactivating specified pathogens using UV (ultraviolet) radiation, exposing invertebrates grown or to be grown in aquaculture to the specified UV-inactivated pathogens to enhance an immune reaction of the exposed invertebrates toward the specified pathogens, and growing the exposed invertebrates in aquaculture. For example, the methods were demonstrated to increase the immune response of oysters to ostreid herpesvirus 1 (OsHV-1) following their prior exposure to UV-inactivated OsHV-1.

The vaccine and plinabulin can be administered concurrently or separately.

Compositions and methods of enhancing immune responses to enteric pathogens
Vaccine vectors capable of eliciting an immune response to enteric bacteria and methods of using the same are provided. The vaccine vectors include a polynucleotide encoding a PAL polypeptide. The PAL polypeptide may be expressed on the surface of the vaccine vector. The vaccine vector may also include a second polypeptide encoding an immunostimulatory polypeptide such as a CD154 polypeptide or an HMGB1 polypeptide.
