GLYCOTARGETING THERAPEUTICS
Several embodiments of the present disclosure relate to glycotargeting therapeutics that are useful in the treatment of transplant rejection, autoimmune disease, food allergy, and immune response against a therapeutic agent. In several embodiments, the compositions are configured to target the liver and deliver antigens to which tolerance is desired. Methods and uses of the compositions for induction of immune tolerance are also disclosed herein.
CARRIER PROTEIN WITH SITE-DIRECTED MUTATION AND USE THEREOF IN PREPARATION OF VACCINE
The present invention relates to a carrier protein with site-directed mutation and use thereof in preparation of a vaccine, wherein the carrier protein is selected from fusion proteins formed by one, two or more of diphtheria toxoid, a non-toxic mutant of diphtheria toxin, a bacterial outer membrane protein and a bacterially expressed protein, wherein an amino acid at at least one site on the carrier protein is mutated into an unnatural amino acid, and the unnatural amino acid contains an azido or alkynyl terminal group. In a mutual reaction process of the carrier protein with site-directed mutation of the present invention and a polysaccharide antigen, a covalent bond is formed, and meanwhile a formed conjugate is in a bead-string state, so that the carrier protein and the polysaccharide antigen can be effectively prevented from being excessively crosslinked. Further, particle size distribution of the conjugate is significantly uniform and controllable, which provides an effective means for improving quality of a polysaccharide-protein conjugate vaccine.
Anti-infection and anti-tumor mucosal immune preparation
The present invention relates to an anti-infection and anti-tumor mucosal immune preparation. The mucosal immune preparation includes mucosal immune substances that are mainly formed by organically bonding polyinosinic-polycytidylic acid, non-antibiotic amino compounds and metal cations through chemical bonds. The present invention provides a slow release effect on a local part or the whole body, prevents the degradation of serum ribonucleases of human beings and primates, prolong the half-life period of the mucosal immune preparation, increases the availability and effectiveness of drugs. The mucosal immune preparation can facilitate the mucosal immunity of the body by mucosal immunity and thus facilitate the activation and proliferation of various immune cells, rather than merely acting on diseased local parts, so that the purposes of anti-infection and anti-tumor prevention and treatment with almost no side effects are realized. Mucosal immunity also avoids the pain of repeated injection so that good compliance is achieved.
NOVEL IMMUNOGENS AND METHODS FOR DISCOVERY AND SCREENING THEREOF
The present application is generally directed to methods for identifying immunogens from organisms and pathogens, and in particular for identifying immunogens which when administered as vaccines elicit a cellular and/or humoral immune response. The present application is also directed to pneumococcal T-cell immunogens, and vaccine compositions comprising one or a combination of pneumococcal immunogens and methods for treating or preventing pneumococcal infections using the vaccines thereof. The present invention also encompasses use of the pneumococcal immunogens for diagnostic purposes to identify a subject with a pneumococcal infection.
DETOXIFIED LIPOPOLYSACCHARIDES (LPS), NATURALLY NON-TOXIC LPS, AND USES THEREOF
An enriched population of modified lipopolysaccharide (LPS) molecular species being: devoid of phosphate group at position C1 of the reducing end of their lipid A domain; and substituted at position C6′ of the non-reducing end of their lipid A domain by a hydrophilic moiety, with the proviso that the hydrophilic moiety is not a hydroxyl group. Also, compositions that include the enriched population of modified LPS and uses of naturally-occurring LPS molecular species and/or enriched population of modified LPS molecular species for treating and/or preventing cancer, inflammatory diseases or infectious diseases, and for stimulating an immune response or vaccinating a subject.
PNEUMOCOCCAL CAPSULAR SACCHARIDE CONJUGATE VACCINE
The present invention is in the field of pneumococcal capsular saccharide conjugate vaccines. Specifically, an immunogenic composition for infants is provided comprising a multivalent Streptococcus pneumoniae vaccine comprising 2 or more capsular saccharide conjugates from different serotypes, wherein the composition comprises a serotype 22F saccharide conjugate. Such a vaccine may be used in infant populations to reduce the incidence of elderly pneumococcal disease such as exacerbations of COPD and/or IPD.
MODIFIED VIRUS-LIKE PARTICLES OF CMV
The present invention relates to virus-like particles of plant virus Cucumber Mosaic Virus (CMV), and in particular to modified VLPs of CMV comprising Th cell epitopes, in particular universal Th cell epitopes. Furthermore, these modified VLPs serve as, preferably, vaccine platform, for generating immune responses, in particular antibody responses, against antigens linked to said modified VLPs. The presence of the Th cell epitopes, in particular universal Th cell epitopes, led to a further increase in the generated immune response.
Compositions and methods for inducing immune tolerance
Several embodiments provided in the present disclosure relate to compositions that carry an antigen to which tolerance is desired, the antigen being coupled, bound, or otherwise joined to a targeting moiety, the targeting moiety configured to direct the composition to the liver of a subject. In several embodiments, the antigen in coupled to the targeting moiety by way of a polymeric linker. In several embodiments, the polymeric linker is configured to liberate the antigen in vivo. Methods of using the compositions to reduce and/or prevent unwanted immune responses against an antigen of interest are also provided.

The invention provides an immunogenic composition comprising staphylococcal antigens, containing protein antigens and conjugates of capsular polysaccharides, n particular Hla, ClfA, SpA and conjugates of capsular polysaccharides. Adjuvanted formulations are also provided. The invention may find use in the prevention and treatment of staphylococcal infections, in particular S. aureus infection and disease.
