Blood-brain barrier permeable peptide compositions that contain variable antigen binding domains from camelid and/or shark heavy-chain only single-domain antibodies are described. The variable antigen binding domains of the peptide compositions bind to therapeutic and diagnostic biomarkers in the central nervous system, such as the amyloid-beta peptide biomarker for Alzheimer's disease. The peptide compositions contain constant domains from human IgG, camelid IgG, and/or shark IgNAR. The peptide compositions include heavy-chain only single-domain antibodies and compositions with one or more variable antigen binding domain bound to one or more constant domains.

Multi-Core Magnetic Metal Oxide Nanoparticles

A method (100) for synthesising multi-core magnetic metal oxide nanoparticles is disclosed. The method comprises providing a first precursor mixture (102) comprising a first metal-containing precursor, a first solvent, and a first nanoparticle clustering agent, and heating the first precursor mixture to thermally decompose the first metal-containing precursor to produce a nanoparticle mixture (104) comprising multi-core magnetic metal oxide nanoparticles. The method further comprises performing at least one seeded growth step (106), each comprising a feeding step in which a further precursor mixture (108) is added to the nanoparticle mixture, the further precursor mixture comprising a further metal-containing precursor and a further solvent, and a heating step in which the nanoparticle mixture is heated to thermally decompose the second metal-containing precursor to achieve growth of the multi-core magnetic metal oxide nanoparticles.

METHOD TO ASSESS NODAL DISEASE USING IRON OXIDE NANOPARTICLES IN MAGNETIC RESONANCE IMAGING

This invention relates to a method to assess nodal disease by magnetic resonance imaging with a contrast agent. The contrast agent comprises biofunctional magnetic nanoparticles coated with polymers outside the iron core and conjugated with antibodies specific to the tumor type to be assessed. Images produced from this magnetic resonance imaging with a contrast agent show heterogenous hypo intensity or heterogenous architecture in regions where binding by the antibodies is present.

MRI based molecular imaging is strongly supported by the accurate quantification of contrast agents. According to an exemplary embodiment of the present invention, contrast agent is applied on the basis of a multiple injection application scheme, during which changes in relaxation rate are determined. This may provide for an accurate determination of rumor vascularity via MRI relaxometry.

PHARMACEUTICAL COMPOSITION AND METHODS FOR USING THE SAME

A pharmaceutical composition includes a plurality of metal nanoparticles and at least one therapeutic agent. Each of the metal nanoparticles includes a core and a stabilizing agent coated on a surface of the core. The at least one therapeutic agent is attached to the stabilizing agent of the metal nanoparticles. Each of the therapeutic agent is an amphiphilic compound and has at least one hydrophobic chain interacting with the stabilizing agent. The pharmaceutical composition may further include a polymer shell encapsulating the metal nanoparticles and the therapeutic agent for enabling controlled release of the therapeutic agent. The pharmaceutical compositions are bifunctional and may be used for diagnosing and treating cancer. Methods for using the pharmaceutical compositions in conjunction with radiation therapy to diagnose and treat cancer are also provided.
