The apparatus has a container with a containment volume with a tissue retention volume and a filtrate volume separated by a filter and with a pellet well for collecting concentrate product in the form of a pellet phase from centrifuge processing. The pellet well is accessible only from above when the apparatus is in an access orientation. Collected pellet phase material may be removed from the pellet well by direct aspiration, without suspending the material in a suspension liquid within the container. Access ports may be configured for access only from above the container. The apparatus may include a tissue collector disposed in the disuse retention volume to engage and collect collagen or other stringy tissue. A method of processing adipose tissue to concentrate leuko stromal vascular cells includes multi-step processing using a portable container.

The aspiration pump operates to continually supply harvested AT to the second flexible tubing while the reinjection pumps causes continuous or pulsed deposition of the AT at injection site. To ensure that internal pressure and/or flow of the AT through a channel of delivery of the AT to the reinjection site does not exceed a predetermined value, the system contains an external pressure sensor configured to measure such internal pressure in absence of a part that is in direct contact with the AT.

A closed system for harvesting fat through liposuction, concentrating the aspirate so obtained, and then re-injecting the concentrated fat into a patient comprises as its main components a low pressure cannula having between about 7 to 12 side holes of about 1-2 mm by 2.0 to 4.0 mm, a spring loaded syringe holder with a constant force or coiled ribbon spring to apply a substantially constant pressure over the fill excursion of the plunger, and a preferably flexible collection bag which is also preferably graduated, cylindrical over most of its body and funnel shaped at its bottom, all of which are connected through flexible tubings to a multi-port valve. The multi-port valve has two flutter/duck bill valves which restrict the fluid flow to a one way direction which effectively allows the syringe to be used to pump fat out of a patient and into a collection bag in a continuous manner. After the bags are centrifuged to concentrate the fat, the excess fluids are separated and the valve is re-connected to permit the syringe pump to reverse fluid flow to graft the concentrated fat back into the patient.

The present disclosure provides an inventive method of treating deteriorated skin tissue in the face or selected areas of the body, wherein the skin tissue has deteriorated due to aging and/or other factors, by injecting mesenchymal stem cells (MSCs) into these areas, resulting in improving and/or restoring the affected areas. In one aspect, the MSCs are obtained autogenously. In another aspect, the MSCs are minimally manipulated. In yet another aspect, treatment of the skin tissue is carried out using bone marrow concentrate that contains the MSCs. A platelet rich plasma (PRP) may be added to a bone marrow concentrate containing MSCs.

The apparatus has a mixing device for mixing contents within the tissue retention volume, and including a rotatable shaft extending from outside of the internal containment volume to inside of the internal containment volume and comprises at least one mixing member that is disposed in the tissue retention volume and is connected with the shaft to move through the tissue retention volume relative to the filter when the shaft is rotated. Methods include use of the apparatus to process human biological material containing adipose, such as to prepare a stromal vascular fraction concentrate product or a fat graft composition.
