The present invention relates to a nutritional composition for improving the temperament of young children. The nutritional composition for an infant or toddler comprises a2-β-casein, wherein the a2-13-casein represents more than 50%, preferably, more than 70%, more preferably, more than 90%, even more preferably, 95%, or most preferably 100% of total β-caseins. The composition has been clinically shown to improve the temperament in toddlers and young children.

The MP-D can significantly improve the colonic pathologic morphology and decrease a colonic histopathologic score and an expression level of colonic interferon-γ (IFN-γ) in an inflammatory bowel disease (IBD) mouse model, and shows the ability to interfere with the occurrence of IBD in mice.

The method firstly employs ultrasonic pretreatment of casein and β-lactoglobulin, followed by enzymatic hydrolysis with a protease to prepare casein and ρ-lactoglobulin polypeptide, and traces the activity of the polypeptide by simulating gastrointestinal digestion, and then simulates absorption by intestinal epithelial cells with Caco-2 cells, to characterize a highly active milk protein polypeptide digested by the gastrointestinal tract and absorbed by the Caco-2 cells simulating absorption by the inner wall of the small intestine. The method has identified five such highly active milk protein polypeptides.
