Methods, systems and solid compositions for reagent delivery
Methods, systems, and compositions featuring a solid, dissolvable reagent composition for delivering the reagent, such as an antibody, probe, chromogen, etc., to a sample. The present invention also features methods of producing said compositions, and automated systems featuring the use of the solid, dissolvable reagent compositions. The solid, dissolvable reagent composition may comprise a water-soluble polymer film, such as a polyvinyl alcohol film, infused with the reagent, wherein when applied to the sample, the water-soluble polymer film with reagent contacts the sample (e.g., tissue) and dissolves.

A sample image capturing system includes a sample holding apparatus configured to hold a sample slide on which a sample film is applied; an imaging apparatus configured to capture the sample on the sample slide; a sample appearance image obtaining apparatus configured to obtain a sample appearance image, where the sample appearance image includes at least an appearance image of the sample film; and a controller configured to: obtain the sample appearance image, identify an appearance characteristic of the sample film based on the sample appearance image, determine a capturing parameter based on the appearance characteristic, and control the imaging apparatus to capture, with the capturing parameter, sample components on the sample slide. The disclosure further relates to a sample image capturing method and a computer-readable storage medium. The disclosure can use a characteristic of the appearance image to achieve accurate capturing of sample images.

Information useful for diagnosis or treatment is acquired by quantitatively evaluating the expression levels of each of a plurality of mutually different domains in the same protein in a human tissue slice, by providing: a staining process of using a plurality of staining reagents having mutually different colorings to stain the plurality of different domains in the same protein; a quantifying step of quantifying the expression levels of each of the plurality of domains; and a calculating step of calculating the ratios between the expression levels of the plurality of domains.

ANTIGEN-COUPLED IMMUNOREAGENTS
The present disclosure provides high-performance immunoreagents for use in a variety of immunologic assays and other related techniques. The immunoreagents comprise a primary antibody and a bridging antigen, wherein the bridging antigen is recognized by a detectable secondary antibody with high affinity. Also provided are compositions comprising panels of immunoreagents specific for multiple different target antigens and compositions comprising pairs of primary immunoreagents and their complementary detectable secondary antibodies. The paired primary immunoreagents and secondary antibodies are useful in a variety of immunologic assays, particularly in highly multiplexed assays, where the structure of the bridging antigen is varied in tandem with variation in the detectable secondary antibody, such that a multiplicity of immunoreagents are provided that are capable of simultaneously detecting a multiplicity of target antigens in a single assay. Also provided are kits comprising the immunoreagents, methods of immunologic assay using the immunoreagents of the disclosure, and methods of preparation of the immunoreagents.
