A method for obtaining dissectates from a microscopic sample using a laser microdissection system having a laser unit includes: a) at least partially circumcising and releasing from the sample dissectate regions of the sample as the dissectates using laser pulses provided by the laser unit; b) transferring the dissectates, by being released from the sample, along dissectate trajectories into receptacles of a dissectate collection unit; and c) positioning the receptacles of a dissectate collection unit using a positioning unit to collect the dissectates. The positioning of the receptacles of the dissectate collection unit using the positioning unit is automatically performed based on estimates of the dissectate trajectories, the estimates of the dissectate trajectories being obtained in a learning mode, the learning mode including obtaining dissectates by repeatedly performing at least steps a) and b). Parameters of the dissectate trajectories are determined for a plurality of dissectates.

The tubular element further comprises a central portion having an aperture defined therein for admitting an aerosol generated by laser ablation from a material sample into the carrier fluid flow when this sample is positioned outside the tubular element at a distance in the range of 0 μm to 100 μm from the aperture.

Controlled separation of laser ablation sample gas for direction to multiple analytic detectors

A laser-ablation-based analytical system can include a sample chamber input, a make-up gas input, a vacuum pump, and an output flow. The sample chamber input can be configured to deliver a sample chamber gas flow comprised of combination of a laser-ablated sample and a sample-carrier gas from a sample chamber. The make-up gas input can be configured to provide an amount of make-up gas to supplement the combination of the laser-ablated sample and the sample-carrier gas. The vacuum pump can be fluidly connected to the sample chamber input and the make-up gas input, the vacuum pump configured to create a negative pressure in a sample transport gas downstream of the vacuum pump, the sample transport gas including the make-up gas, the laser-ablated sample, and the sample-carrier gas. The output flow can be configured to deliver the sample transport gas from the vacuum pump to a detection device.

LASER ABLATION SAMPLING SYSTEM AND METHOD

A method and system for sampling a solid sample material can include the step of mounting the sample material on a support. A sample surface is coated with a surface treatment composition in a dry deposition process. A solvent supply conduit for supplying solvent to the sample surface and a solvent exhaust conduit for withdrawing solvent from the sample surface can be provided. Solvent is flowed from the solvent supply conduit to the surface treatment composition and the sample surface such that the solvent contacts the surface treatment composition. A laser beam is directed from a laser source to the sample and the surface treatment composition. The laser beam will ablate the sample and the surface treatment composition in portions intersected by the laser beam. Ablated sample material enters the solvent liquid and will be transported with the solvent away from the sample surface through the solvent exhaust conduit.

Disclosed herein are systems and methods for performing imaging mass cytometry, including analysis of labelling atoms by elemental (e.g., atomic) mass spectrometry. Aspects include a sampling system having, and method of using, a femtosecond (fs) laser and/or laser scanning. Alternatively or in addition, aspects include systems and methods for co-registering other imaging modalities with imaging mass cytometry.
