SINGLE CELL SELECTION AND ISOLATION

According to one method, a sample with cells contains a phototagging agent. The sample is imaged to identify at least one target cell to be isolated. The identified target cell in the sample is selectively irradiated with photo-activating light for selectively activating the phototagging agent in the target cell to change its fluorescence response. The irradiated target cell is isolated from other cells in the sample based on a difference in its fluorescence response compared to non-activated phototagging agent in the other cells. Further aspects are directed to a corresponding microscope system and chemical compound for use as the phototagging agent.

In some embodiments, the method includes: receiving the electrical signals from the detector; mathematically combining individual decay curves in the electrical signals into a supercurve, the supercurve comprising a number of components, each component having a time constant and a relative contribution to the supercurve; and quantifying a relative contribution of each component to the supercurve.

Disclosed are systems are methods for identifying the composition of single aerosol particles, particularly that of bioaerosol particles. A continuous timing laser tightly coupled with a pulse ionization laser is used to index aerosol particles, measure particle properties, and trigger the ionization laser to fire when each particle enters the beam of the trigger laser. Ionized fragments and optionally photons produced when each particle is struck by the ionization laser are analyzed using one or more detectors including a TOF-MS detector and an optical detector. Individual single particle spectra are aligned and denoised prior to averaging.
