Methods, compositions and systems for microfluidic assays

Provided herein, among other aspects, are methods and apparatuses for analyzing particles in a sample. In some aspects, the particles can be analytes, cells, nucleic acids, or proteins and contacted with a tag, partitioned into aliquots, detected by a ranking device, and isolated. The methods and apparatuses provided herein may include a microfluidic chip. In some aspects, the methods and apparatuses may be used to quantify rare particles in a sample, such as cancer cells and other rare cells for disease diagnosis, prognosis, or treatment.

INTEGRATED PARTICULATE MATTER SENSOR WITH CAVITY

A particulate matter (PM) sensor comprises a substrate forming a cavity (5), the substrate comprising a semiconductor chip (4), and a light source (1) arranged in the cavity (5). The light source (1) is adapted to emit a light beam (7). The light beam (7) forms a detection volume (8) for particulate matter (9) outside the cavity (5). Optionally, the particulate matter sensor comprises an optical element (2) delimiting the cavity (5) at one end. The optical element (2) is configured to shape the light beam (7). Further, the particulate matter sensor comprises at least one photodetector (3) that is integrated into a surface of the semiconductor chip (4). The surface into which the at least one photodetector (3) is integrated faces the detection volume (8). The at least one photodetector (3) is adapted to detect light (10) scattered by particulate matter (9) in the detection volume (8).

A combined capability sensor includes a first laser source and a second laser source. The first laser source is configured to emit light having a wavelength in at least one of an infrared spectrum and a visible spectrum and the second laser source is configured to emit light having a wavelength in a blue or ultraviolet spectrum. A director is configured to direct the first and second laser source to a detection zone. A first sensor is configured to detect scattered light originating from the first laser source, thereby detecting a presence of smoke in the detection zone. A second sensor is configured to detect fluoresced light originating from the second laser source, thereby detecting a presence of a biological agent in the detection zone.

The invention encompasses analyzers and analyzer systems that include a single molecule analyzer, methods of using the analyzer and analyzer systems to analyze samples, either for single molecules or for molecular complexes. The single molecule uses electromagnetic radiation that is translated through the sample to detect the presence or absence of a single molecule. The single molecule analyzer provided herein is useful for diagnostics because the analyzer detects single molecules with zero carryover between samples.

A method for detecting biomarkers with shortened test time and enhanced precision is provided. A sample from the body fluid is made to flow over a sensor surface coated with a capture antibody to allow binding of a biomarker in the sample to the capture body. An optical method detects and counts the individual binding events along the sensor surface with single molecule resolution, and difference in the binding events along the sensor surface is detected in real-time and analyzed to determine the biomarker concentration.

Methods and systems for capture object-based assays, including for determining a measure of the concentration of an analyte molecule or particle in a fluid sample, are described. The methods and systems may relate to high sensitivity detection of analytes, sometimes using assay conditions and sample handling that result in the capture and detection of a high percentage of the analyte molecules or particles in a fluid sample using relatively few capture objects. Apparatuses and methods for immobilizing capture objects with respect to assay sites, in some instances with unexpectedly high efficiencies are also described. Some such apparatuses involve the use of force fields and fluid meniscus forces, alone or in combination, to facilitate or improve capture object immobilization. Also described are techniques for utilizing a relatively high percentage of capture objects in an assay sample, such as by using disclosed sample washing techniques, imaging systems, and analysis procedures that can reduce capture object loss.
