HOLOGRAPHIC RECONSTRUCTION APPARATUS AND METHOD
Provided are an improved holographic reconstruction apparatus and method. A holographic reconstruction method includes: obtaining an object hologram of a measurement target object; extracting reference light information from the obtained object hologram; calculating a wavenumber vector constant of the extracted reference light information, and generating digital reference light by calculating a compensation term of the reference light information by using the calculated wavenumber vector constant; extracting curvature aberration information from the object hologram, and then generating digital curvature in which a curvature aberration is compensated for; calculating a compensated object hologram by multiplying the compensation term of the reference light information by the obtained object hologram; extracting phase information of the compensated object hologram; and reconstructing 3-dimensional (3D) shape information and quantitative thickness information of the measurement target object by calculating the quantitative thickness information of the measurement target object by using the extracted phase information of the compensated object hologram.

A microscope device (1) comprises an illumination optical system (10,20) for illuminating a sample, a detection optical system (40) for receiving light from the sample, a detector (60) for detecting the light from the sample via the detection optical system and outputting a detection signal of the light, a data processor (66) for generating at least one of a three-dimensional refractive index distribution and a two-dimensional phase distribution in the sample based on the detection signal output from the detector, and a modulation element group that is provided at a position of a pupil or a position conjugate with the pupil in only the illumination optical system, and has light transmittance changing continuously within a surface of the pupil or within a surface conjugate with the pupil.

An observation apparatus including: a light-source that emits illumination light and excitation light upward from below a specimen; and an image-capturing optical system having an objective lens that focuses, below the specimen, transmitted light, which is the illumination light that is emitted from the light-source, that is reflected above the specimen, and that has passed through the specimen, and fluorescence that is generated in the specimen that has been irradiated with the excitation light emitted from the light-source, wherein the light-source is disposed radially outside the objective lens.
LIGHT SHEET MICROSCOPE
A light sheet microscope includes: a detection objective configured to image a target region of a sample located in a focal plane of the detection objective; an illumination objective configured to focus an illumination light beam in the sample, the detection objective and the illumination objective being opposite to one another, and the optical axis of the detection objective and the optical axis of the illumination objective being parallel to one another; a first light deflection device having at least one first deflection surface and one second deflection surface, which are each arranged offset to the optical axis of the detection objective and are configured to deflect the illumination light beam focused by the illumination objective in a direction perpendicular to the optical axis of the detection objective such that a deflected illumination light beam forms a light-sheet-like illumination light distribution focused in the focal plane.
