The present disclosure provides improved systems and methods for automated cell identification/classification. More particularly, the present disclosure provides advantageous systems and methods for automated cell identification/classification using shearing interferometry with a digital holographic microscope. The present disclosure provides for a compact, low-cost, and field-portable 3D printed system for automatic cell identification/classification using a common path shearing interferometry with digital holographic microscopy. This system has demonstrated good results for sickle cell disease identification with human blood cells. The present disclosure provides that a robust, low cost cell identification/classification system based on shearing interferometry can be used for accurate cell identification. For example, by combining both the static features of the cell along with information on the cell motility, classification can be performed to determine the type of cell present in addition to the state of the cell (e.g., diseased vs. healthy).

The electromagnetic radiation assembly is further configured to diffract the received electromagnetic radiation and transmit a diffracted electromagnetic radiation. An image capture assembly is configured to capture an image of the diffracted electromagnetic radiation and produce the hologram of the cross-section of the object from the captured image. The hologram of the cross-section includes information regarding a single cross-section of the object.

INCOHERENT HOLOGRAPHIC IMAGING WITH METASURFACES

A birefringent metasurface lens formed by an array of nanoposts generates different phase profiles for different incident electromagnetic waves of different polarization. A first polarization is focused at a first focal length, while a second polarization is focused at a second focal length. A variable phase retarder and a polarizer generate interference patterns for each phase difference between the two incident polarizations. The interference patterns are used to generate a hologram, allowing reconstruction of the image of an object.

The present disclosure discloses a hologram recording device and a hologram recording method. The hologram recording device comprises an object imaging unit comprising a cavity for accommodating an object whose interior wall is a reflective surface, and a light modulating unit configured to produce incident light and reference light interfering with the incident light and to direct the incident light to the object imaging unit. The cavity is provided with an imaging aperture for imaging of the object and at least one light incidence aperture for allowing the incident light to enter the cavity and irradiate on the object, such that an image of the object is formed at a location corresponding to the imaging aperture outside the cavity, and image light produced upon the imaging of the object interferes with the reference light at the location for recording of an image surface hologram.
