The present disclosure relates to improved holographic reconstruction device and a method. In one aspect, the present disclosure relates to improved holographic reconstruction device and method that can measure a digital hologram regardless of optical characteristics of an object to be measured, by an all-in-one type system integrating a transmissive system that measures an object transmitting light and a reflective system that measures an object reflecting light.
System for spatial multiplexing
Some embodiments are directed to a technique having an off-axis interferometric geometry that is capable of spatially multiplexing at least six complex wavefronts, while using the same number of camera pixels typically needed for a single off-axis hologram encoding a single complex wavefront. Each of the at least six parallel complex wavefronts is encoded into an off-axis hologram with a different fringe orientation, and all complex wavefronts can be fully reconstructed. This technique is especially useful for highly dynamic samples, as it allows the acquisition of at least six complex wavefronts simultaneously, optimizing the amount of information that can be acquired in a single camera exposure. The off-axis multiplexing holographic system of some embodiments provide an off-axis holography modality that is more camera spatial bandwidth efficient than on-axis holography. Moreover, the off-axis interferometric system allows simple simultaneous acquisition of at least six holographic channels, making it attractive for imaging dynamics.

Provided are on-axis and off-axis digital hologram generating device and method. The on-axis and off-axis digital hologram generating device includes an object phase generator configured to access a phase file of an object stored in a storage device and generate object phase information from the phase file of the object; a digital object light generator configured to generate digital object light information based on a light property of object light input by a user and the object phase information generated by the object phase generator; a digital reference light generator configured to generate digital reference light information based on a light property of reference light input by the user; and a digital hologram generator configured to generate a digital hologram based on hologram property information input by the user, the digital object light information generated by the digital object light generator, and the digital reference light information generated by the digital reference light generator.
Virtual staining of cells in digital holographic microscopy images using general adversarial networks
A cell visualization system includes a digital holographic microscopy (DHM) device, a training device, and a virtual staining device. The DHM device produces DHM images of cells and the virtual staining device colorizes the DHM images based on an algorithm generated by the training device using generative adversarial networks and unpaired training data. A computer-implemented method for producing a virtually stained DHM image includes acquiring an image conversion algorithm which was trained using the generative adversarial networks, receiving a DHM image with depictions of one or more cells and virtually staining the DHM image by processing the DHM image using the image conversion algorithm. The virtually stained DHM image includes digital colorization of the one or more cells to imitate the appearance of a corresponding actually stained cell.

INTERFEROMETRIC SCATTERING CORRELATION (ISCORR) MICROSCOPY
A method of characterizing one or more particles in a fluid, e.g. a liquid, using interferometric scattering optical (iSCAT) microscopy. The method involves illuminating a region of a fluid using an objective lens so that light is scattered by one or more particles in the fluid. The scattered light and reference light are captured using the objective lens and interfere at an imaging device. A succession of images of the interference is processed to determine image correlation values which define a gradual decorrelation over time from which a property of the particle(s) is determined.
Method and system for recording digital holograms of larger objects in non-laboratory environment
A method for optimally producing a holographic image using a Holographic Optical Element (HOE) and the HOE meant for controlling directions and divergences of light beams to impart system compactness. The system uses concave and convex lenses and other beam expanding, splitting, modulating and combining optics for realization of compactness and high throughput. The thin laser beam is split using a holographic optical element and a conventional beam splitter. A neutral density filter adjusts the intensity of a reference beam to match the intensity of an object beam so that high quality digital holograms can be recorded. Effects of vibrations are minimized by the compact optical design, by anti-vibration mounts, by mounting all the opto-mechanical components on a single rigid platform and by enclosing the system. An electro-optical sensor array records holograms digitally and an algorithm numerically reconstructs and further quantifies the results using a personal computer/laptop/tablet etc.
DARK FIELD DIGITAL HOLOGRAPHIC MICROSCOPE AND ASSOCIATED METROLOGY METHOD
A dark field digital holographic microscope and associated metrology method is disclosed which is configured to determine a characteristic of interest of a structure. The dark field digital holographic microscope includes an illumination branch for providing illumination radiation to illuminate the structure; a detection arrangement for capturing object radiation resulting from diffraction of the illumination radiation by the structure; and a reference branch for providing reference radiation for interfering with the object radiation to obtain an image of an interference pattern formed by the illumination radiation and reference radiation. The reference branch has an optical element operable to vary a characteristic of the reference radiation so as to reduce and/or minimize variation in a contrast metric of the image within a field of view of the dark field digital holographic microscope at a detector plane.

An image processing apparatus includes an acquisition unit that acquires a hologram obtained by imaging a plurality of granules contained within an imaging visual field, a generation unit that generates, from the hologram, phase difference images at positions different from each other in an optical axis direction in a case in which the hologram is captured, a specifying unit that specifies a plurality of image ranges in a direction of a plane intersecting the optical axis direction, which correspond to the plurality of granules, in an averaged image obtained by averaging at least some of the phase difference images, and an extraction unit that extracts the phase difference image at a center position of a corresponding granule in the optical axis direction for each of the plurality of image ranges.
