Container for blood volume assessment
Disclosed are methods, materials and devices for approximation of blood volume in a fluid, such as in a biological fluid collected during a surgical procedure. The method and devices include the use of a RBC flocculant, such as polyDADMAC, and an approximate blood hematocrit for the type of animal, as well as a calculated RBC packing ratio corresponding to the collection device being used. Also provided is a Blood Indicator Panel (BIP), comprising a series of markings calculated from an observed red blood settlement volume, the average animal type hematocrit, and a calculated RBC packing ratio “η” value for the collection device. Pediatric (about 200 ml or 250 ml size container), adult human (about 1,000 ml-1,500 ml) and veterinary (about 500 ml-2,500 ml) collection containers are also disclosed, that include a RBC flocculant, for use in approximating blood volume in a fluid.
Agricultural sampling system and related methods
A coulter assembly for collecting soil samples from an agricultural field in one embodiment comprises an annular collection blade configured for penetrating soil to capture a sample, an annular cam ring configured for stationary mounting to a frame of an agricultural vehicle and comprising a cam track, a blade hub coupled to the blade for rotatably supporting the annular collection blade from the annular cam ring, and a movable sample collector mounted to the annular collection blade. The moveable sample collector is configured and operable for extracting a soil sample as the annular collection blade is rotated through the soil. The moveable sample collector in one embodiment comprises a piston mechanism including a cylinder and rod movably disposed therein which actuated by the annular cam ring to alternatingly open and close a collection end of the cylinder as the annular collection blade rotates through the soil.
Determining concentration of oil in water by extraction
A method is implemented to determine a concentration of oil in a water sample. A specified amount of cyclohexane is added to the water sample to form a mixture. The mixture is stirred. An oil phase is extracted from the mixture. An absorbance value of the extracted oil phase is measured at a specified wavelength in the visible light spectrum. The specified wavelength is in a range of from 390 nanometers (nm) to 600 nm. The concentration of oil in the water sample is determined based on the measured absorbance value of the extracted oil phase.
ANALYTICAL ULTRACENTRIFUGATION FOR CHARACTERIZATION OF RECOMBINANT VIRAL PARTICLES
Provided herein are methods to characterize preparations of recombinant viral particles using analytical ultracentrifugation. Recombinant viral particles include recombinant adeno-associated viral particles, recombinant adenoviral particles, recombinant lentiviral particles and recombinant herpes simplex virus particles. Variant species of recombinant viral particles including empty capsids and recombinant viral particles with variant genomes (e.g., truncated genomes, aggregates, recombinants) can be identified and quantitated. The methods can be used to characterize preparations of recombinant viral particles regardless of the sequence of the recombinant viral genome or the serotype of the recombinant viral capsid.
Interface Detection And Control Using A Photodetector Array
Systems and methods are provided for determining and controlling the location of an interface between separated fluid components within a channel of a centrifugal separation chamber being rotated about a rotational axis. Light from a light source is received by a collimator, which directs collimated light through the channel in a direction substantially parallel to the rotational axis. At least a portion of the collimated light exiting the channel is received by a light detector configured as a photodetector array. A signal emitted by the light detector is received by a controller, which determines the location of an interface between separated fluid components within the channel based at least in part on the signal. When the controller determines that the interface is not at a target location within the channel, the controller controls a centrifugal separator and/or a pump system to move the interface to the target location.
DETERMINATION OF TOTAL CRUDE OIL IN WATER BY ABSORBANCE SPECTROPHOTOMETRY
A method for quantifying a crude oil in water is provided. The method includes selecting an ultraviolet/visible (UV/Vis) wavelength to perform a measurement, preparing calibration solutions in xylene, and preparing a calibration curve from the calibration solutions. A sample is prepared including extracting the crude oil from the water in a two-phase separation with xylene. An absorbance of the sample in the xylene is measured at the UV/Vis wavelength. A concentration of the crude oil in the water is calculated from the absorbance.

ISOLATION TUBE
A separation container for extracting a portion of a sample for use or testing and method for preparing samples for downstream use or testing are provided. The separation container may include a body defining an internal chamber. The body may define an opening, and the body may be configured to receive the sample within the internal chamber. The separation container may further include a seal disposed across the opening, such that the seal may be configured to seal the opening of the body, and a plunger movably disposed at least partially inside the internal chamber. The plunger may be configured to be actuated to open the seal and express the portion of the sample.
