A hyper-productive chromatography technique includes providing a scalable and stackable chromatographic cassette, loading a sample to be processed, operating the scalable chromatographic cassette having an adsorptive chromatographic bed having a volume greater than 0.5 liter by establishing a flow at a linear velocity greater than 500 cm/hr with a residence time of the loading step of less than one minute.
Method for storage and stabilization of a target substance
The invention relates to a system and method for the stable storage of sensitive biological or chemical target substance, in a bound form on certain capture media. The method comprised providing a sample containing the target substance in a suitable buffer; combining the sample with a capture media to effect reversible binding of the target substance to the capture media; and storing the capture media with the target substance at between about −20 and 20° C.; and recovering the target substance from the capture media. The target substance recovered maintains the desired activity. Also provides are methods for reducing aggregates in the sensitive biological or chemical target substance.
Chromatography column support
Herein is reported the use of a chromatography column support comprising at least one plane of symmetry, one axis of symmetry, at least three legs, at least three straight connectors, whereby the connectors define a plane that is perpendicular to the axis of symmetry of the support, whereby the connectors are connected to each other at the axis of symmetry, whereby each leg is connected to a connector, whereby each leg is perpendicular to the plane defined by the connectors, whereby all legs are on the same side of the plane defined by the connectors for stabilizing the packing of a chromatography column.
MULTIPLE MEMBRANE SEPARATION CAPSULE
A membrane capsule for biological and chemical separations comprising a cassette comprising an upper surface and a lower surface adjoined by a cassette sidewall, an inlet and an outlet located on the upper and lower surfaces of the cassette, tubes fluidly connected to the inlet and the outlet, holes or slots in the tubes to facilitate separation, and a membrane wrapped, pleated, and/or spiral wound around each of the tubes. Methods of separation comprising flowing fluid flow through the inlet of the membrane capsule, allowing the fluid to permeate through the holes or slots of the tubes, separating biological and/or non-biological substances, collecting the fluid within a reservoir, and draining fluid from the reservoir.

Longitudinal gradient chromatography columns
An ion exchange chromatography column contains an ion exchange stationary phase that includes a charged substrate, a plurality of first particles, and a plurality of second particles. The plurality of first particles each include first ion exchange groups and the first particles are ionically bound to the charged substrate. The plurality of second particles each include second ion exchange groups and the second particles are ionically bound to the charged substrate. The first particles having a first ion exchange group density, and the second particles having a second ion exchange group density. The first ion exchange group density is greater than the second ion exchange group density. The ion exchange chromatography column has a number of zones connected in series where each zone can have a varying level of first ion exchange groups and second ion exchange group from the inlet zone to the outlet zone.

A cyclic chromatographic purification method for the isolation of a product from a feed mixture consisting of the product and at least one further component representing impurities, which impurities bind stronger to the chromatographic stationary phase than the product is given. The method uses at least two chromatographic adsorbers as chromatographic stationary phase, grouped into only one first adsorber section (1) and one second adsorber section (2), wherein if an adsorber section comprises more than one chromatographic adsorber these are permanently connected in series, wherein the first adsorber section (1) has a first adsorber section inlet and a first adsorber section outlet, and the second adsorber section (1) has a second adsorber section inlet and a second adsorber section outlet.
Stackable planar adsorptive devices
Adsorptive bed devices include a monolithic scaffold having a stress absorbing rigid structure and open cells filled with adsorptive beads. The monolithic scaffold restricts movement of the plurality of adsorptive beads, absorbs stress induced by a hydraulic pressure gradient along a direction of liquid flow. In one embodiment the adsorptive bed is packed into a chromatography column, and in another embodiment the adsorptive bed is sealed in a monolithic block.
