Analyzers and methods for making and using analyzers are described such as a method in which multiple absorption readings of a liquid assay are obtained by a photodetector using multiple light sources having at least three separate and independent wavelength ranges and with each of the absorption readings taken at a separate instant of time. Using at least one processor and calibration information of the liquid assay, an amount of at least two analytes within the liquid assay using the multiple absorption readings is determined.

A host antigen-specific antibody testing system and method. The a ternary complex of the antigen, a ligand-bound anti-host IgM, and a non-host anti-antigen IgG detector conjugate selectively form a quaternary complex with host antibodies, wherein the host antibodies and IgG compete for the antigen, and the anti-host IgM binds the host antibodies. The quaternary complex is retained by an immobilized IgM ligand binding agent, and any residual ternary complex is retained by a later encountered immobilized anti-non-host IgG. If sufficient host antibodies have a high affinity for the antigen, the complex is detected at the quaternary complex detection region based on the presence of the detector, and if there are insufficient high affinity host antibodies, the ternary complex migrates past the quaternary complex detection region and is retained and detected at a control region.

A magnet is adjacent the microfluidic chip and is movable relative to the microfluidic chip. A valve control is capable of actuating certain ones of the controllable blocking valves in response to a control signal.

COMPOSITIONS AND METHODS FOR DETECTING AUTOANTIBODIES
The present invention relates to the field of autoimmunity. More specifically, the present invention provides compositions and methods useful for detecting autoantibodies. In one embodiment, a method for detecting autoantibodies to ZnT8 comprises the steps of (a) contacting in a first mixture a biological sample obtained from a patient with a ZnT8-antibody complex, wherein the ZnT8-antibody complex comprises ZnT8 and at least one detectably labeled anti-ZnT8 antibody or antigen-binding fragment thereof that specifically binds to the cytoplasmic domain of ZnT8; (b) contacting in a second mixture the first mixture of step (a) with an immunoglobulin G (IgG) labeled with a tag molecule; (c) contacting the second mixture of step (b) with a solid substrate coated with a capture molecule that specifically binds the tag molecule; and (d) detecting a signal emitted from the detectably labeled anti-ZnT8 antibody or antigen-binding fragment thereof.
