Chromatography device and method for filtering a solute from a fluid
A chromatography device for removing a solute from a fluid is provided. The device has a first plate having an inlet and a first channel. The first channel directs the fluid from the inlet towards chromatographic media housed in a chamber coupled to the first plate. The chamber has a leading edge for receiving the fluid from the first channel and a trailing edge for delivering the fluid to a second channel. The chromatographic media is configured to remove the solute from the fluid as the fluid passes through the chamber. The device also has a second plate coupled to the chamber having the second channel and an outlet. The second channel directs the fluid from the chamber to the outlet. The direction of flow of fluid through the first channel and the second channel is transverse to a direction of flow of the fluid through the chromatographic media. A method of removing a solute from a fluid is also provided.
Laterally-fed membrane chromatography device
A method of forming a frame around a membrane stack for a laterally-fed membrane chromatography device is provided. The method includes placing a membrane stack having one or more membrane layers on a bottom surface of body of a master mold, the body having opposed side walls and opposed end walls, the opposed side walls spaced apart by a distance greater than a length of the membrane stack, the opposed end walls spaced apart by a distance greater than a width of the membrane stack; placing a cap on the body of the master mold to enclose the membrane stack in the master mold, the cap having at least one opening for injecting a material into a space defined by the end walls of the master mold, the side walls of the master mold, end walls of the membrane stack side walls of the membrane stack, the bottom surface of the body and an inner surface of the cap; injecting the material into the space around the membrane stack; and curing the material to form a frame around the membrane stack.
CHROMATOGRAPHIC CASSETTE
A chromatographic cassette includes a cassette including a chamber, chromatographic media disposed within the cassette chamber, a distribution network fluidly coupled to the chromatographic media and an inlet port and an outlet port coupled to the distribution network. A hyper-productive chromatography technique includes providing a scalable and stackable chromatographic cassette, loading a sample to be processed, operating the scalable chromatographic cassette having an adsorptive chromatographic bed having a volume greater than 0.5 liter by establishing a flow at a linear velocity greater than 500 cm/hr with a residence time of the loading step of less than one minute.
Systems and methods of preparing stabilized lipid assemblies
Direct polymerization of lipid monomers or polymer scaffolding of non-lipid monomers coupled with irradiation or redox polymerization performed at neutral pH resulted in stabilized lipid assemblies. An initiator-buffer component and NaHS03 redox mixture polymerizes reactive lipid monomers at near neutral pH conditions to preserve functionality of reconstituted membrane proteins. Improved stability of black lipid membranes (BLMs) is attained by chemical cross-linking of polymerizable, hydrophobic and commercially available non-lipid monomers partitioned into the suspended lipid membranes, and by suspending the BLMs across low surface energy apertures. Substrate apertures having low surface energy modifiers with amphiphobic properties facilitated a reproducible formation of BLMs by promoting interactions between the lipid tail and the substrate material. In addition, polymeric lipid bilayer membranes were prepared by photochemical or redox initiated polymerization of polymerizable lipid monomers, and disposed onto supporting substrates for use in chromatography columns.
PACKING ASSEMBLY FOR CHROMATOGRAPHY MEDIA AND METHODS OF USE THEREOF
A packing assembly for forming a packed-bed for a chromatography device is described herein. The packing assembly includes a bottom plate and a first top plate. The first top plate has a top plate hole centrally positioned therein. The packing assembly also includes a middle plate. The middle plate has a middle plate hole centrally positioned therein. The middle plate hole is aligned with the top plate hole when the packing assembly is in an assembled state. The packing assembly also includes a second top plate. The second top plate has a protrusion extending outwardly. At least a portion of the protrusion has a same size and a same shape as the top plate hole to be received in the top plate hole when the packing assembly is in a packing state. Methods of packing chromatography media in a packing assembly and assembling a chromatography device are also described herein.
STACKABLE PLANAR ADSORPTIVE DEVICES
Adsorptive bed devices include a monolithic scaffold having a stress absorbing rigid structure and open cells filled with adsorptive beads. The monolithic scaffold restricts movement of the plurality of adsorptive beads, absorbs stress induced by a hydraulic pressure gradient along a direction of liquid flow. In one embodiment the adsorptive bed is packed into a chromatography column, and in another embodiment the adsorptive bed is sealed in a monolithic block. In another embodiment, the adsorptive bed device includes an adsorptive block, first and second planar distributors and peripheral seal.
Stackable planar adsorptive devices
A lattice and distribution network for a stackable chromatography cassette comprising: a peripheral seal; at least one screen forming the lattice surrounded by the peripheral seal, each at least one screen comprising a plurality of struts in a latticed arrangement; a first internal distribution network fluidly coupled to the lattice and surrounded by the peripheral seal; a second internal distribution network disposed opposite the first internal distribution network, fluidly coupled to the lattice and surrounded by the peripheral seal; wherein a direction of fluid flow is established from the first internal distribution network through the lattice to the second internal distribution network; and wherein preferential streamlines are minimized.

LATERALLY-FED MEMBRANE CHROMATOGRAPHY DEVICE
A laterally-fed membrane chromatography device for removing a solute from a fluid is provided. The device has a top plate, a middle plate and a bottom plate. The top plate has an inlet and a top channel for directing the fluid from the inlet towards a membrane stack. The middle plate houses the membrane stack. The membrane stack has a leading edge for receiving the fluid from the top channel, a trailing edge for distributing the fluid to the bottom channel, and is configured to remove the solute from the fluid as the fluid passes through the membrane stack. The bottom plate has a bottom channel and an outlet. The bottom channel is for directing the fluid from the membrane stack to the outlet. The top channel directs the fluid form the inlet over the leading edge in a direction that is transverse to the direction of flow of the fluid through the membrane stack.
