PROCESS FOR FORMING MICROBUBBLES WITH HIGH OXYGEN CONTENT AND USES THEREOF

Formulations containing a carrier and microbubbles encapsulating oxygen gas, and methods for making and using the formulations are described herein. The formulations are manufactured by a process which includes high shear homogenization. The resulting microbubble suspension may be centrifuged to further concentrate the microbubbles. The resulting concentrated LOM suspension preferably has an oxygen content ranging from 50 to 99% (vol). Prior to administration to a patient, the viscosity of the LOM suspension may be reduced to the desired viscosity, preferably similar to the viscosity of the patient's blood. The resulting LOM formulation typically has an oxygen concentration ranging from 65 to 80% (vol). The microbubbles are formed from one or more lipids, preferably one or more phospholipids, most preferably DSPC, and preferably also contain one or more stabilizing agents/excipients, preferably cholesterol.

A method for normalizing blood vessels of lesions is disclosed, which includes administering an effective amount of oxygen-loaded microbubbles to a subject in need by intravenous injection, and projecting ultrasound from a ultrasonic emission device into the lesions for rupturing the oxygen-loaded microbubbles and releasing the oxygen to the lesions. Each of the oxygen-loaded microbubbles comprises oxygen and a water insoluble gas, and the particle size of microbubbles is 0.520 m.

The present invention relates to a polymer solution for use as a rheological blood substitute. The rheological blood replacement solution comprises in its basic composition a water-soluble cellulose polymer with a molecular weight of 100 kDa or greater, polyethylene oxide and a solvent. The rheological blood replacement solution of the invention can be used in a variety of applications, e.g. as blood substitute, calibration agent or reference agent. The blood replacement solution is based on a polymer solution, which can also be used as pharmaceutical composition, e.g. for the prevention or treatment of haemorrhage or shock-related disorders.

The present invention concerns a multipart fluid system for dialysis therapy, wherein the multipart fluid system comprises an anticoagulation fluid and at least one treatment fluid from the group consisting of dialysis fluid and infusion fluids. According to the invention the anticoagulation fluid comprises 15-40 mM citrate; and the at least one treatment fluid comprises 1.5-8 mM citrate and 10 mM bicarbonate. The present invention further concerns a system for citrate anticoagulation in an extracorporeal blood circuit.
