Controlled release dosage forms are described herein. The controlled release formulations described herein provide prolonged delivery of high dose drugs that are highly water soluble and highly hygroscopic. In specific embodiments, controlled release dosage forms for delivery of a drug selected from GHB and pharmaceutically acceptable salts, hydrates, tautomers, solvates and complexes of GHB. The controlled release dosage forms described herein may incorporate both controlled release and immediate release formulations in a single unit dosage form.

Room-temperature stable dosage forms for oral delivery of calcitonin are disclosed herein. Dosage forms for oral delivery of calcitonin with low water content are also disclosed herein. Further disclosed herein are methods for producing such room-temperature stable dosage forms and low-water content dosage forms. Methods of treatment comprising the administration of such room-temperature stable dosage forms and low-water content dosage forms are also disclosed herein.

A tamper resistant drug dosage is described. The drug dosage form includes a matrix polymer, a scaffold polymer, and a therapeutic agent, and the porosity of the drug dosage form is less than 10%. Methods for making and using the tamper resistant drug dosage forms are described.

Thrombin-inhibiting acylated pyrazole-pyridone compounds of formula (II) are disclosed herein, as well as pharmaceutical compositions, including tablets, that contain acylated pyrazole-pyridone compounds. Provided is a device for granulating, agglomerating, pelletising, drying and/or coating, the device including a swirl chamber, a distribution chamber, wherein the swirl chamber is separated from the distribution chamber by a base and wherein a powder to be granulated or a powder mixture to be granulated is presented in the swirl chamber, the device further including at least one agitator for thoroughly mixing the powder to be granulated or the powder mixture to be granulated and at least one addition device for a liquid, wherein the base is designed in several parts and at least one base part is horizontally and/or vertically displaceable, with the result that that the base becomes a distributor plate. Also provided is a method for granulating, agglomerating, pelletising, drying and/or coating using such a device and a base suitable for use as a distributor plate in a convective drying apparatus.
