TREATMENT OF SKIN DISORDERS WITH TOPICAL COMPOSITIONS COMPRISING TAPINAROF AND A PDE4 INHIBITOR

Provided herein is a topical combination composition comprising tapinarof, a PDE4 inhibitor and optionally at least one additional active agent selected from a retinoid, benzoyl peroxide (BPO), a Janus kinase inhibitor (JAK inhibitor), a corticosteroid of potency class 1-4, an acaricide and combinations thereof. The active agents in the composition of this invention are in encapsulated or non-encapsulated form, according to need. The above compositions are useful for the treatment, prevention or alleviation of a skin disorder selected from psoriasis, dermatitis, acne, rosacea, ichthyosis, scaling skin, imbalance of skin barrier and tinea and exhibit synergistic and/or additive effects allowing to reduce the active agents amounts in the combination compositions.

An oral amphetamine extended release liquid suspension is described. The compositions contain a combination of an uncoated amphetamine-cation exchange resin complex, a barrier coated amphetamine-cation exchange resin complex-matrix, and an uncomplexed amphetamine, wherein one or more of these components contains blends of different forms of amphetamines. Either the modified release coated and/or the uncoated amphetamine-cation exchange resin complex may have two forms of amphetamine in a complex with a single cation exchange resin. Following administration of a single dose of the composition, a therapeutically effective amount of amphetamine is reached by about one hour and the composition provides at least a thirteen hour effect post-dose.

The invention relates to an extract of Ashwagandha that exhibit enhanced bioactivity and bioavailability comprising of enriched withanolide glycosides and saponins; with negligible amount of alkaloids, withanolide aglycones and oligosaccharides. The extract as disclosed prepared from root, stems, leaves and whole plant of Ashwagandha further shows improved immunomodulatory activity, anti-inflammatory activity, anti stress activity, antidiabetic activity and sleep quality. The disclosure also provides a method of improving bioactivity of withanolide glycosides even at lower doses, by the administration of an enteric coated formulation of extract of Ashwagandha to humans. The enteric coating protects the composition from hydrolysis in the acidic environment of the stomach to release the withanolide glycoside in neutral/ alkaline pH in gastrointestinal tract (GIT) thus enhancing the absorption. Further the process of preparation of the extract of Ashwagandha enriched with withanolide glycosides and saponins are disclosed along with various formulations.
