Collagen compositions and uses for biomaterial implants

Compositions containing purified collagen biomaterial derived from tissues, for example, insoluble amnion, soluble amnion, soluble chorion of the human placenta, are provided. The collagen compositions can be used to promote wound healing, promote tissue regeneration, prevent or reduce scarring, reduce local inflammation, minimize tissue rejection, promote graft integration. Methods for using the collagen composition as a biomaterial implant for dermal filling, skin grafting, and hair transplantation are also provided.

The present invention discloses a braided silk scaffold with adjustable mechanical and degradation properties, and a preparation method and use thereof, belonging to the field of three-dimensional scaffold materials for tendon/ligament repair. The preparation method includes braiding at least one silk strand to form a silk core; placing 1-6 bundles of silk cores in a braiding machine, and braiding at least one layer of silk cladding on the surface of the silk cores to form a silk base frame; removing sericin from the silk base frame; soaking the silk base frame in a collagen solution with a concentration of 3-20 mg/ml, and cross-linking the silk base frame in a vacuum thermal cross-linking machine to obtain the silk scaffold. The braided silk scaffold with adjustable mechanical and degradation properties according to the present invention has good mechanical properties and biocompatibility.

A tissue construct is provided that comprises a pancreas derived microvessel fragment and a pancreatic islet cell. The pancreas derived microvessel fragment and the pancreatic islet cell can be incorporated into a biocompatible medium. Tissue constructs can be comprised of other cells, such as stem cells, combined with the pancreas derived microvascular fragment. Methods for isolating microvessel fragments from a pancreas are also provided and include enzymatic digestion of pancreatic tissue and separation of microvessel fragments from endocrine and exocrine tissue. Methods for treating diabetes are further provided and include administration of the tissue constructs.

Disclosed are devices and methods for transplanting cells, such as pancreatic endoderm cells, into a host. The devices include a non-woven fabric external to a cell-excluding membrane, and the non-woven fabric and/or cell-excluding membrane can be perforated. Treatment of the host with immunosuppressive reagents, required to inhibit allograft rejection due to perforations in the cell delivery device, does not compromise maturation or function of transplanted pancreatic endoderm cells.
