SYSTEM FOR ELECTROSPRAY IONIZATION WITH INTEGRATED LC COLUMN AND ELECTROSPRAY EMITTER

An integrated system for liquid separation and electrospray ionization includes an emitter-enabled capillary column including an emitter portion and a column portion; and a retractable protective sleeve for covering and/or supporting the emitter portion along at least a portion of its axis. The protective sleeve is mounted around the emitter-enabled capillary column. The retractable protective sleeve is moveable to an extended position wherein a tip of the emitter portion is covered by the protective sleeve. A resilient member is provided to bias the protective sleeve towards the extended position. The protective sleeve is enclosed and moveable within an outer electrically conductive sheath adapted for insertion within a holder having a high-voltage contact point. The sheath is adapted to contact the high-voltage contact point and provide an electrical connection to enable the emitter-enabled capillary column to receive a high voltage. The sheath has a recess to receive the high-voltage contact point.

Systems and methods for use in introducing samples to an analytical device for single particle compositional analysis. Suitable analytical devices include, for example, an inductively coupled plasma-optical emission spectrometer. Prior to introduction to the analytical device, the sample gas is exchanged with argon gas, for example, using a gas exchange device. The analytical device may be calibrated with a liquid sample which is aerosolized prior to entry into the analytical device.

METHOD FOR MEASURING OR IDENTIFYING A COMPONENT OF INTEREST IN SPECIMENS

A method for identifying a target component of interest in a specimen by probe electrospray ionization mass spectrometry, including: (1) adsorbing the target component of interest onto an extraction phase for adsorbing the target component of interest from the specimen by immersing a probe into the specimen, wherein the probe is at least partially coated with the extraction phase; (2) removing the probe from the specimen and optionally quick rising it by dipping in or spraying water or acetone/water mixtures; (3) adhering solvent to the extraction phase; (4) desorbing the target component of interest into the solvent from the extraction phase; (5) electrospraying the target component of interest desorbed in the solvent adhered to the probe on the ionization source at atmospheric pressure by applying a voltage to the probe to spray aerosolized ionized droplets out of the probe; and (6) identifying the target component of interest present in the aerosolized ionized droplets.

The present disclosure relates to an apparatus and method to achieve electrospray ionization at femtoliter/minute to nanoliter/minute flow rates including relatively rapid alternation between such flow rates within the same device. These flow rates provide enhanced and relatively more uniform ionization of sprayed compounds for subsequent analytical evaluations.

Presented herein are apparatuses for use in capillary separations. An apparatus includes a coupling that integrates a capillary with a voltage source, a sheath liquid system, a fluid exit port, and a manifold. The coupling may be an elbow connector or equivalent. The manifold receives incident light from an incident light input, and emitted light is collected by a collected light output. The capillary enters the manifold at an input for the capillary, traverses the coupling, and terminates at the fluid exit port, for example an electrospray emitter. The capillary may also enter the manifold at an input for the capillary and terminates inside the manifold.

SPRAY CHAMBERS AND METHODS OF USING THEM

Devices, systems and methods including a spray chamber are described. In certain examples, the spray chamber may be configured with an outer chamber configured to provide tangential gas flows. In other instances, an inner tube can be positioned within the outer chamber and may comprise a plurality of microchannels. In some examples, the outer chamber may comprise dual gas inlet ports. In some instances, the spray chamber may be configured to provide tangential gas flow and laminar gas flows to prevent droplet formation on surfaces of the spray chamber. Optical emission devices, optical absorption devices and mass spectrometers using the spray chamber are also described.

MICROFLUIDIC DEVICES WITH GAS CHANNELS FOR SAMPLE NEBULIZATION

Methods, devices, and systems for performing nebulization of a sample from a fluid channel of a microfluidic device are described. The systems or devices disclosed herein may comprise microfluidic devices that comprise a gas channel used for nebulization of the sample at a fluid outlet of the microfluidic device. In some instances, the disclosed devices may be designed to perform isoelectric focusing followed by further characterization of the separated analytes using electrospray ionization coupled with nebulization to introduce the samples into a mass spectrometer. The disclosed methods, devices, and systems provide for fast, accurate separation and characterization of protein analyte mixtures or other biological molecules by isoelectric point.
