Methods and compounds are disclosed for wound healing by modulating autophagy. A formulation for modulating autophagy comprises a first modulating compound (FAM) selected from compounds having the general structure (I): ##STR00001## wherein: L represents a linker selected from —C≡C—, (a tolan), —CH═CH— (a stilbene, preferably trans); or —CR.sub.a═CR.sub.b— a stilbene derivative; where R.sub.a and R.sub.b are independently H or phenyl optionally substituted with —(R.sup.3).sub.p or —(R.sup.4).sub.q; R.sup.1 to R.sup.4 are independent substituents at any available position of the phenyl rings, preferably at 3, 3′, 4, 4′, and/or 5, 5′; and m, n, p, and q are independently 0, 1, 2, or 3 representing the number of substituents of the rings, respectively, but at least one of m or n must be ≥1. Each R.sup.1 to R.sup.2 is independently selected from substituents described herein, including but not limited to hydroxyl, alkoxy, halo, halomethyl and glycosides. The formulation may also include an auxiliary autophagy modulating compound (AAM) as described herein. The formulation may include a hydrogel formed by the compounds themselves or otherwise and may include salts and/or complexes.

The present invention relates generally to methods of use and compositions useful to reduce dark spots, age spots, and/or unwanted pigmentation of skin. The composition includes a combination of niacinamide, phytic acid, Rosmarinus officinalis leaf extract, Chondrus crispus extract, and a combination of Saxifraga sarmentosa extract, Carica papaya (papaya) fruit extract, and Psidium guajava (guava) fruit extract. The composition is effective to reduce dark spots, age spots, and/or unwanted pigmentation of skin. The composition is effective to inhibit tyrosinase and/or Peroxisome Proliferator-Activated Receptor Gamma (PPAR-γ) activity. The composition is further effective to reduce melanin content in skin, particularly dark skin, in some instances by at least 30%.

Disclosed herein are skin care compositions, comprising vitamin E (e.g., alpha-tocopherol), a thiourea derivative of histidine (e.g., ergothioneine), a vitamin B3 compound (e.g., niacinamide) and/or a pyrrolidone carboxylic acid and/or a salt thereof (e.g., sodium PCA) as well as to methods of using these compositions.

A method of dechlorinating skin, especially dechlorinating skin, hair and/or nails of humans including contacting said skin, hair and/or nails with an aqueous composition for sufficient time to reduce halogen effects on the skin, hair and/or nails. The aqueous composition comprising water and an oil-soluble bis-ascorbate in a weight ratio of about 0.1 to 25% by weight of the total aqueous composition, a surfactant, and a buffering agent maintaining pH of the aqueous composition in anaerobic conditions at between 7.8 and 8.3 at room temperature for at least 10 days. Optionally there is 0.05% to 30% by weight of suspended carbon particles.

Compositions and methods for reducing the symptoms of skin conditions associated with the nail and surrounding tissue are provided. A medicated pad for cleaning and treating the nail and surrounding tissue is described, wherein said pad is saturated with a therapeutic composition comprising certain botanicals, an antimicrobial agent, a surfactant, and a moisturizing agent. The skin and nail conditions can include fungal infections of the nails. Kits for treating fungal infections of the nails are likewise described. Optionally, the kit may include a hydrating composition which includes shea butter, aloe, vitamins, arnica, emu oil, urea, and Vitamins C, A, E, and D. A bandage comprising a therapeutic pad instilled with an antifungal agent is also provided.
