Each input may have several forms such as base, hover, engaged, completed, and error. User input is anticipated. The base form of the anticipated input cursor is displayed to prompt the user for the anticipated input. If user hover is detected that matches anticipated input, the hover form is displayed to confirm the match to the user. If user input is detected that matches anticipated input, the engaged form is displayed as confirmation. If user input is completed that matches anticipated input, the completed form is displayed as confirmation. If user hover or input does not match anticipated input, the error form is displayed to indicate mismatch. Not all cursors must have all forms, and some cursors may have multiples of some forms.

The method further comprises preparing a regenerative component of the autologous composition comprising: collecting blood from the mammal; delivering the blood to a tube in the presence of about 4% citric acid; centrifuging the blood to separate a platelet-rich plasma component from a whole blood; collecting the platelet-rich plasma component; and mixing the supernatant component with the platelet-rich plasma component to provide the autologous composition. Disruption of neuronal elements in the disk includes destroying nerve endings without substantially affecting the central body of the nerve, suppressing activation of the nerve endings, and inhibiting the growth of nerve endings into the disk. Disruption of vascular elements includes causing the vascular extensions to retract from the disk, or suppressing the formation of such extensions. The therapeutic agent may be administered locally via an interbody pump, a bolus or a depot, or may be administered systemically.

The methods include administering to the subject a therapeutically effective amount of multipotent stem cells, wherein each multipotent stem cell is transformed with a recombinant nucleic acid molecule comprising a heterologous promoter operably linked to a nucleic acid encoding platelet derived growth factor (PDGF) B, and wherein the multipotent stem cells express a sufficient amount of PDGFB to increase bone mass and strength or bone fracture healing. A lentiviral vector also is disclosed that includes a phosphoglycerate kinase-1 (PGK) promoter operably linked to a nucleic acid encoding PDGFB.
