ULTRAFINE IRON OXIDE NANOPARTICLE-BASED MAGNETIC RESONANCE IMAGING T1 CONTRAST AGENT
Provided is a T1 contrast agent for magnetic resonance imaging. The T1 contrast agent includes fine iron oxide nanoparticle cores and micelles encapsulating the core particles. The micelles include a nonionic surfactant consisting of a hydrophilic moiety containing at least two chains and a hydrophobic moiety containing at least one C.sub.10-C.sub.30 hydrocarbon chain. The T1 contrast agent of the present invention is a novel one based on fine iron oxide nanoparticles that can replace conventional gadolinium-based T1 contrast agents. The T1 contrast agent based on fine iron oxide nanoparticles according to the present invention is harmless to humans, is rapidly distributed in the blood, and has a uniform size, ensuring its uniform contrast effect. In addition, the T1 contrast agent of the present invention enables image observation for at least 1 hour to up to 2 hours and is excreted through the kidneys and liver. Therefore, the T1 contrast agent of the present invention avoids the problems encountered in conventional gadolinium-based contrast agents.
Melanocortin 1 receptor ligands and methods of use
The subject invention pertains to a modified MC1R peptide ligand comprising a peptide that is a melanocortin 1 receptor (MC1R) ligand and a functionality or linker, such as a click functionality, for conjugation to a surface or agent. The modified MC1R peptide ligand can be coupled, e.g., via a click reaction with a complementary click functionality attached, to a moiety to form an MC1R-targeted agent. Drugs, contrast agents, polymers, particles, micelles, surfaces of larger structures, or other moieties can be targeted to the MC1R. The subject invention also pertains to a MC1R peptide ligand-micelle complex comprising a peptide that is a melanocortin 1 receptor ligand connected via a click reaction product to a micelle. The micelle is stable in vivo and can target melanoma tumor cells by association of the peptide ligand with the MC1R or the tumor and selectively provide a detectable and/or therapeutic agent (such as an imageable contrast agent and/or anti-cancer agent) selectively to the tumor cell.
Porphyrin modified telodendrimers
The present invention provides amphiphilic telodendrimers that aggregate to form nanocarriers characterized by a hydrophobic core and a hydrophilic exterior. The nanocarrier core may include amphiphilic functionality such as cholic acid or cholic acid derivatives, and the exterior may include branched or linear poly(ethylene glycol) segments. Nanocarrier cargo such as hydrophobic drugs and other materials may be sequester in the core via non-covalent means or may be covalently bound to the telodendrimer building blocks. Telodendrimer structure may be tailored to alter loading properties, interactions with materials such as biological membranes, and other characteristics.

Polyphosphazene delivery system for inorganic nanocrystals
Nanoclusters comprising inorganic nanocrystals and a biodegradable polymer are disclosed. The inorganic nanocrystals have a mean particle size of 1 to 500 nm. The inorganic nanocrystals are contained within a core of the nanoclusters, on the surface of the nanoclusters, contained within a core of the nanoclusters, dispersed throughout the nanoclusters, or a combination thereof. The biodegradable polymer allows the inorganic nanocrystals to be excreted renally over a period of time. The nanoclusters can be used for medical imaging or other biomedical applications.
AMPHIPHILIC POLYMER NANO MICELLE CONTAINING POLY-3,4-DIHYDROXYPHENYLALANINE CHELATED FERRIC IONS
The disclosure discloses an amphiphilic polymer nano micelle containing poly-3,4-dihydroxyphenylalanine chelated ferric ions, in which ferric ions are chelated with a catechol structure on a side chain of a biodegradable poly-3,4-dihydroxyphenylalanine block. The disclosure also provides a method for preparing the above micelle, comprising: complexing an amphiphilic polymer containing poly-3,4-dihydroxyphenylalanine with a ferric ion compound, and obtaining the amphiphilic polymer nano micelle containing poly-3,4-dihydroxyphenylalanine chelated ferric ions through a solvent replacement method. The micelle prepared by the disclosure is used as a Fe.sup.3+ magnetic resonance Ti imaging contrast agent, which can avoid toxic or side effects caused by a traditional gadolinium reagent, has a longitudinal relaxation rate of 5.6 mM.sup.1.Math.s.sup.1, can cycle for 150 min in a mice body, and has an obvious imaging effect and a far higher comprehensive performance than that of a commercial gadolinium contrast agent, and as well as a promising application prospect.
Spherical rHDLs For Targeted Imaging
A new approach to targeting imaging agents to macrophage-rich sites of interest is disclosed. Compositions of the invention are rHDL and HDL-like liposomal compositions, protein constituents of which, apolipoproteins A-I and/or A-II or fragments thereof are used not only as structural but also as targeting agents. This is achieved by certain controlled chemical or enzymatic modification of apolipoproteins A-I or A-II or fragments thereof. Such modification converts these apolipoproteins to substrates for macrophage scavenger receptors and results in the improvement of contrast agent-(HDL/modified apolipoprotein)-particle association with macrophages and/or absorption (uptake) by macrophages when compared to that of the contrast agent-(HDL/apolipoprotein)-particle constructed with non-modified naturally occurring apo A-I. The compositions can be used for noninvasive specific in vivo molecular detection and localization of macrophage-rich sites of interest using imaging techniques such as computed tomography (CT), gamma-scintigraphy, positron emission tomography (PET), single photon emission computed tomography (SPECT), magnetic resonance imaging (MRI).
Nanoparticle compositions for generation of regulatory T cells and treatment of autoimmune diseases and other chronic inflammatory conditions
The present invention relates to nanoparticles for the targeted delivery of antigen to liver cells, in particular, liver sinusoidal endothelial cells (LSEC) and/or Kupffer cells, and for the in vivo generation of regulatory T cells, notably CD4+CD25+FOXP3+ regulatory T cells (Treg). The invention provides pharmaceutical compositions and methods for the prevention and treatment of autoimmune diseases, allergies or other chronic inflammatory conditions, and for generation of regulatory T cells. The nanoparticles used in the invention comprise a) a micelle comprising an amphiphilic polymer rendering the nanoparticle water-soluble, and b) a peptide comprising at least one T cell epitope associated with the outside of the micelle. The micelle may or may not comprise a solid hydrophobic core.
