20220266212 · 2022-08-25 · ·

The present disclosure provides fluidic devices and fluidic device assemblies, including microfluidic devices and cartridges comprising the same, that in illustrative embodiments, can be used to make particles or protein precipitates, or to monitor precipitate formation. The fluidic devices typically include channels that connect a reaction well to an inlet port and an outlet port, and a fluidic constriction channel that is configured to help retain fluids in the reaction well and/or promote mixing within the reaction well. In some aspect, fluidic devices are interconnected into fluidic assemblies that can be used in continuous process methods.

The present invention provides methods for generating a library of synthetic polynucleotides. The present invention also provides methods for generating proteins encoded by the library of synthetic polynucleotides. In addition, provided herein are methods for determining the soluble expression of said proteins. This invention is based, in part, on the discovery of a method for selecting optimal oligonucleotides in combination with performing a phosphorylation reaction, ligation reaction and PCR amplification in a single reaction vessel to produce synthetic polynucleotides in a multiplex manner.

ENHANCED COMPOSITE LIQUID CELL (CLC) DEVICES, AND METHODS FOR USING THE SAME

Enhanced composite liquid cell (CLC) devices and methods of using the same are provided. The devices find use in, among other applications, CLC mediated nucleic acid library generation protocols, e.g., for use in next generation sequencing applications.

MULTI-PLANE MICROARRAYS

An array including a solid support having a plurality of contours along its exterior surface. A first subset of contours is positioned along the exterior surface of the solid support to form a first pattern of features and a second subset of contours is positioned along the exterior surface to form a second pattern of features. The contours of the first subset are juxtaposed with the second subset along the exterior surface, whereby the first and second patterns form an interleaved pattern. The features of the first pattern occur at a first elevation z.sub.1 and the features of the second pattern occur at a second elevation z.sub.2. The features of the first pattern are configured to attach analytes at a different elevation relative to analytes attached to the features of the second pattern.

System and method with fiducials in non-rectilinear layouts

Fiducial markers are provided on a patterned array of the type that may be used for molecular analysis, such as sequencing. The fiducial markers may have configurations and layouts that enhance their detection in image or detection data, that facilitate or improve processing, that provide encoding of useful information, and so forth. Examples of the fiducial markers may include non-rectilinear layouts that may provide for more robust location of both the fiducial markers and sites of the patterned array.

Method of immobilizing a nucleic acid probe to a solid support

A nucleic acid probe, a method of immobilizing the nucleic acid probe to a solid support and the solid support including the immobilized probes using UV light. The nucleic acid probe includes a terminus anchor chain portion, and a capture portion wherein the terminus anchor chain portion includes a sequence of at least 18 nucleotides composed of stretches of up to 5 nucleotides of base type X with intermediate nucleotide(s) of base type Cytosine (C) and optionally one nucleotide of base type Guanine (G) or a sequence with at least 90% similarity thereto, wherein each base type X independently of each other designate base type Thymine (T) or base type Uracil (U).

COMBINATORIAL BARCODE SEQUENCES, AND RELATED SYSTEMS AND METHODS

A kit for use with a nucleic acid sequencing instrument can include a plurality of combinatorial barcodes sequences meeting the following criteria: each of the combinatorial barcode sequences comprise a plurality of iterations of a sequence motif, where the sequence motif comprises a first nucleotide base from a first group of nucleotide bases followed by a second nucleotide base from a second group of nucleotide bases, the first group and the second group differing from each other; and the plurality of combinatorial barcode sequences is at least 1,000,000 different barcode sequences.

Explosive and narcotics detection dog training with vapour or aerosol air impregnation

The present invention provides devices and methods for the impregnation of air with the vapor or aerosol of a substance in a ‘controllable manner to enable the testing or training of detection means to evaluate and quantify the presence of the substance in an enclosed volume, and iη• particular to enable production of training aids and quality assurance test items for use in canine-olfaction based security screening.

Array of micromolded structures for sorting adherent cells

An apparatus for collecting or culturing cells or cell colonies includes: a common substrate formed from a flexible resilient polymeric material and having a plurality of wells formed therein; and a plurality of rigid cell carriers releasably connected to said common substrate, with said carriers arranged in the form of an array, and with each of the carriers resiliently received in one of the wells. A method of collecting or culturing cells or cell colonies with such an apparatus is carried out by depositing a liquid media carrying cells on the apparatus so that said cells settle on or adhere to said the carriers; and then (c) releasing at least one selected carrier having said cells thereon by gradual application of release energy to each carrier from the cavity in which it is received (e.g., by pushing with a probe).
