A sample preparation module accepts a sample including a target analyte. The sample preparation module processes the sample through several reaction chambers and a solid phase column. Different reagents are present in the reaction chambers. The eluted analyte is then transferred to the amplification module, where it is further processed and amplified for optical analysis.

Provided herein are compositions and devices for diagnosing and treating a viral infection in a subject and/or detecting a viral nucleic acid in a sample. In one example, a sample is treated in a first chamber and the sample is flowed into a second chamber, where any pathogenic nucleic acid is detected by oligonucleotides that are specific to the pathogen under test. Further, the oligonucleotides comprise a cleavage site for a restriction enzyme in the second chamber, which cleaves oligonucleotide-pathogenic nucleic acid hybrid resulting in the exposure of an enzyme that was being held by the oligonucleotide to its substrate, which generates a colorimetric and/or another visual readout.

A reaction container includes a transparent base including a resin material and having recessed portions formed in one or more regions of the transparent base such that the recessed portions are recessed from a surface of the transparent base, and a cover member including a thermoplastic resin material and positioned on the transparent base such that the cover member forms a flow path including a gap extending over the recessed portions between the cover member and the surface of the transparent base and has a spacer portion welded by laser irradiation to the transparent base outside the one or more regions of the transparent base. The cover member absorbs infrared light and transmits light having a wavelength within a range of visible light such that the cover member has light transmittance in the range of from 480 nm to 570 nm.

Systems and devices for analysis of samples

Systems and methods for analysis of samples, and in certain embodiments, microfluidic sample analyzers configured to receive a cassette containing a sample therein to perform an analysis of the sample are described. The microfluidic sample analyzers may be used to control fluid flow, mixing, and sample analysis in a variety of microfluidic systems such as microfluidic point-of-care diagnostic platforms. Advantageously, the microfluidic sample analyzers may be, in some embodiments, inexpensive, reduced in size compared to conventional bench top systems, and simple to use. Cassettes that can operate with the sample analyzers are also described.

MEASUREMENT OF AN ANLYTE WITH A CARTRIDGE

A method of performing an optical measurement of an analyte in a processed biological sample using a cartridge is provided. The cartridge is operable for being spun around a rotational axis. The method comprises: placing the biological sample into a sample inlet; controlling the rotational rate of the cartridge to process a biological sample into the processed biological sample using a fluidic structure; controlling the rotational rate of the cartridge to allow the processed biological sample to flow from the measurement structure inlet to an absorbent structure via a chromatographic membrane, and performing an optical measurement of a detection zone on the chromatographic membrane with an optical instrument. An inlet air baffle reduces evaporation of the processed biological sample from the chromatographic membrane during rotation of the cartridge.
