20210252457 · 2021-08-19 · ·

A system, method and device are disclosed for bio-processing a feed stream and providing a constant output by operating a continuous single-pass tangential-flow process. The single-pass process provides high conversion concentration while operating at relatively low feed flow rates, and the process can also be used to provide constant output diafiltration.

SINGLE PASS TANGENTIAL FLOW FILTRATION HYBRID CONFIGURATIONS FOR ENHANCING CONCENTRATION OF MACROMOLECULE SOLUTIONS
20210253633 · 2021-08-19 ·

This disclosure provides a method for concentrating a solution of a macromolecule that is retained on at least two semi-permeable membranes that have different molecular weight cutoffs (MWCOs), the method comprising passing the solution through a hybrid configuration of said semi-permeable membranes staged in series in a single pass tangential flow filtration (SPTFF) apparatus. The method is applicable to the efficient concentration of biological macromolecules such as proteins, antibodies and nucleic acids.

METHOD AND SYSTEM FOR OPERATING A HIGH RECOVERY SEPARATION PROCESS

A turbocharger includes a turbocharger housing having an impeller housing comprising a circular cross-section. A main nozzle is disposed within the turbocharger housing communicating a first portion of a fluid stream to a first volute. A first auxiliary channel and a first auxiliary nozzle communicating a second portion of the fluid stream to the first volute. The first auxiliary nozzle is downstream of the main nozzle. A second auxiliary channel and a second auxiliary nozzle communicate a third portion of the fluid stream to the first volute. The second auxiliary nozzle is downstream of the first auxiliary nozzle. A valve assembly is selectively coupling the first auxiliary channel to the first auxiliary nozzle and the second auxiliary channel to the second auxiliary nozzle.

Single-pass filtration systems and processes

The present invention provides, in various embodiments, hybrid single-pass tangential flow filtration assemblies, disposable single-pass tangential flow filtration assemblies, scalable single-pass tangential flow filtration assemblies and adaptable modular single-pass tangential flow filtration assemblies. In other embodiments, the invention relates to processes for recovering proteins from the surface of a filtration membrane in a single-pass tangential flow filtration assembly and for cleaning a tangential flow filtration assembly. In additional embodiments, the invention provides methods of increasing the processing capacity of a single-pass tangential flow filtration assembly.

Single pass tangential flow filtration systems and tangential flow filtration systems with recirculation of retentate

A method of filtering a liquid feed is described, comprising passing a liquid feed through a single pass tangential flow filtration (SPTFF) system and recovering the retentate and permeate from the system in separate containers. A method of filtering a liquid feed is also described comprising passing a liquid feed through a tangential flow filtration (TFF) system, recovering permeate and a portion of the retentate from the system in separate containers without recirculation through the TFF system, and recirculating the remainder of the retentate through the TFF system at least once. The methods of the invention can be performed using an SPTFF or a TFF system that comprises manifold segments to serialize the flow path of the feed and retentate without requiring diverter plates.

Two-stage gas separation unit contained within a single housing

A two-stage gas-separation membrane system includes two identical membrane modules held within a single casing. A feed gas is directed into the first module, so as to produce permeate and retentate streams. One of the latter streams then becomes the feed gas for the second module, and reaches the second module through a core tube located within the module. The product of the second module is the product gas for the system. The gas streams entering the two modules flow in mutually opposite directions. This arrangement makes it feasible to provide a two-stage system while using only the number of ports that would be needed for a single stage.

A purification system for a colloidal dispersion containing an unwanted dissolved substance in the dispersing liquid phase is described which includes a back-pressure regulating feature, disposed in a first conduit and downstream of a semi-permeable filter element. This is used to force a portion of the unpurified dispersing liquid across the filter element and into a second conduit. The second conduit is associated with an adsorptive element that serves to remove the unwanted dissolved substance from the filtered portion of the dispersing liquid. A mixing chamber is disposed downstream of the back-pressure regulating feature whereby the purified dispersing liquid portion from the second conduit rejoins the bulk flow of the colloidal dispersion passing through the first conduit. In a preferred embodiment, hollow fibers are used which serve as the back-pressure regulating feature, the adsorptive element, and the mixing chamber before being discharged at the outlet of the device.
