A technique relates to a machine for sorting. A removable cartridge includes a nanofluidic module. The removable cartridge includes an input port and at least two output ports. The nanofluidic module is configured to sort particles in a sample fluid. A holder is configured to receive the removable cartridge. A pressurization system is configured to couple to the input port of the removable cartridge. The pressurization system is configured to drive the sample fluid into the nanofluidic module for separation to the at least two output ports.

Each excitation channel has one light source and one single excitation filter and one dichroic mirror. One excitation channel is capable of exciting multiple fluorescent dyes and can be used to distinguish multiple dyes in combination with multiple emission filters. The excitation channels are driven by a motor that can automatically switch the two excitation channels for taking images of up to seven different fluorescent dyes. An algorithm to calibrate the crosstalk between different fluorescent dyes is also provided. Also provided is a method for analyzing digital PCR data using a ratio of two fluorescence emission readings.

The valve is formed in a valve body housing a first path portion, a second path portion, and an coupling zone between the first and second path portions. A shutter is arranged in the coupling zone and has a shutting portion of ferromagnetic material that is deformable under the action of an external magnetic field between an undeformed position, wherein the shutter closes the coupling zone, and a deformed position, wherein the shutter at least partially frees the coupling zone. The shutting portion of the shutter is formed by a rubber membrane incorporating particles, for example of ferrite particles.
