Automated microscopic cell analysis

This disclosure describes single-use test cartridges, cell analyzer apparatus, and methods for automatically performing microscopic cell analysis tasks, such as counting and analyzing blood cells in biological samples. A small measured quantity of a biological sample, such as whole blood, is placed in a mixing bowl on the disposable test cartridge after being inserted into the cell analyzer. The analayzer also deposits a known amount of diluent/stain in the mixing bowl and mixes it with the blood. The analyzer takes a measured amount of the mixture and dispenses in a sample cup on the cartridge in fluid communication with an imaging chamber. The geometry of the imaging chamber is chosen to maintain the uniformity of the mixture, and to prevent cells from crowding or clumping as it is transferred into the imaging chamber by the analyzer. Images of all of the cellular components within the imaging chamber are counted and analyzed to obtain a complete blood count.

Methods for biological sample processing and analysis

Provided are methods for biological sample processing and analysis. A method can comprise providing a substrate configured to rotate. The substrate can comprise an array having immobilized thereto a biological analyte. A solution comprising a plurality of probes may be directed, via centrifugal force, across the substrate during rotation of the substrate, to couple at least one of the plurality of probes with the biological analyte. A detector can be configured to detect a signal from the at least one probe coupled to the biological analyte, thereby analyzing the biological analyte.

Method and apparatus for electroporation of acoustically-aligned cells

Transfer of genetic and other materials to cells is conducted in a hands-free, automated and continuous process that includes flowing the cells between electroporation electrodes to facilitate delivery of a payload into the cells, while acoustophoretically focusing the cells. Also described is a control method for the acoustophoretic focusing of cells that includes detecting locations of cells flowing through a channel, such as with an image analytics system, and modulating a drive signal to an acoustic transducer to change the locations of the cells flowing in the channel. Finally, an electroporation driver module is described that uses a digital to analog converter for generating an electroporation waveform and an amplifier for amplifying the electroporation waveform for application to electroporation electrodes.

A nucleic acid amplification method includes a step of heating a first region of a container housing a droplet containing a target nucleic acid and a sample necessary for amplification of the target nucleic acid to a denaturation temperature of the target nucleic acid and heating a second region different from the first region to a synthesis temperature of the target nucleic acid, and an amplification step of repeating a cycle through a denaturation stage at which the droplet housed in the container is moved to and retained in the first region and a synthesis stage at which the droplet is moved to and retained in the second region at a plurality of times. At the amplification step, periods of part of cycles of the plurality of cycles are made shorter than periods of the other cycles.

Methods and containers are provided for identifying a species, illustratively a bacterial species. Illustrative methods comprise amplifying various genes in the nucleic acid from the bacterial species in a single reaction mixture using pairs of outer first-stage primers designed to hybridize to generally conserved regions of the respective genes to generate a plurality of first-stage amplicons, dividing the reaction mixture into a plurality of second-stage reactions, each using a unique pair of second-stage primers, each pair of second-stage primers specific for a target bacterial species or subset of bacterial species, detecting which of the second-stage reactions amplified, and identifying the bacterial species based on second-stage amplification. Methods for determining antibiotic resistance are also provided, such methods also using first-stage primers for amplifying genes known to affect antibiotic resistance a plurality of the second-stage reactions wherein each pair of second-stage primers specific for a specific gene for conferring antibiotic resistance.

COLUMN-BASED DEVICE AND METHOD FOR RETRIEVAL OF RARE CELLS BASED ON SIZE, AND USES THEREOF

A column-based device and method for retrieving cells of interest were enclosed. The said device comprises a column comprising (i) an inner wall defining an inner chamber with inlet and outlet openings, (ii) a perforated plug disposed adjacent to the outlet opening, (iii) a sleeve insert with a channel and disposed within the chamber and adjacent to the perforated plug, and (iv) a filtering means housed within sleeve insert sandwiched between two sealing means. In particular, Tumor-derived endothelial cell clusters (TECCs) as characterized multiple nuclei, expression of endothelial markers (PECAM1, VWF and CDH5), and non-expression of leukocyte, megakaryocyte and platelets markers, may be retrieved using the disclosed device. Also encompassed are methods, reagents and kits for the diagnosis and prognosis of cancers by detecting for the presence of TECCs isolated from blood samples using the claimed device.

The present invention is, in combination, a bone marrow isolation tube and a centrifuge tube. The combination includes a centrifuge tube which defines a tube cavity and includes an upper cylindrical section connected to a lower tapered section which terminates at a bottom reservoir. The isolation tube is adapted to be received within the centrifuge tube cavity. The isolation tube includes a body with a tubular shaped outer surface wherein the interior of the isolation tube defines a lumen which includes an upper cylindrical lumen section and a lower conical lumen section terminating at an orifice. The isolation tube includes at least one wing extending radially outward from the outer surface of the isolation tube. The wing is configured to make contact with the interior surface of the centrifuge tube and wedge the isolation tube into the tapered section of the centrifuge tube during centrifuging. The present invention is also, in combination, a bone marrow isolation tube and a centrifuge tube. The centrifuge tube defines a tube cavity and includes at least two support members defining an insertion space and attached to the interior surface in an upper cylindrical section, the upper cylindrical section connected to a lower tapered section which terminates at a bottom reservoir. The isolation tube is adapted to be received within the insertion space with a flange that is configured to make contact with the support members of the centrifuge tube thereby wedging the isolation tube into the insertion space of the centrifuge tube during centrifuging.
