METHODS AND COMPOSITIONS FOR DOCKING BIOTINYLATED ANTIGENS ON THE EXTERIOR OF BACTERIAL OUTER MEMBRANE VESICLES
20230083394 · 2023-03-16 ·

The present disclosure is directed to a system for displaying antigens. This system includes an outer membrane vesicle comprising a lipid bilayer and a synthetic antigen receptor comprising an outer membrane scaffold protein fused to a biotin-binding protein, where the outer membrane scaffold protein is incorporated in the lipid bilayer and the biotin-binding protein is displayed outside the outer membrane vesicle. Also disclosed are therapeutic compositions, nucleic acid constructs, expression vectors, and methods of eliciting an immune response in a subject.

HIV-1 Env fusion peptide immunogens and their use

Embodiments of immunogens based on the HIV-1 Env fusion peptide and methods of their use and production are disclosed. Nucleic acid molecules encoding the immunogens are also provided. In several embodiments, the immunogens can be used to generate an immune response to HIV-1 Env in a subject, for example, to treat or prevent an HIV-1 infection in the subject.

MODIFIED VESICULAR STOMATITIS VIRUS GLYCOPROTEIN AND USES THEREOF FOR THE TREATMENT OF BRAIN TUMORS

A vaccine for treating and/or preventing a brain tumor. More particularly, a modified vesicular stomatitis virus glycoprotein (VSV-G) including at least one tumor antigen, or a fragment thereof, for use in preventing and/or treating a brain tumor in an individual in need thereof, when administered before a surgery intended to remove all or part of the tumor, such as, a tumor resection. The inventors have shown that vaccination of individual with a brain tumor with a vaccine including a nucleic acid sequence encoding a modified VSV-G may be combined to a tumor resection in order to ameliorate the prognostic of the individuals.

TRITERPENE SAPONINS, METHODS OF SYNTHESIS AND USES THEREOF

The present invention relates to triterpene glycoside saponin-derived adjuvants, syntheses thereof, intermediates thereto, and uses thereof. QS-7 is a potent immuno-adjuvant that is significantly less toxic than QS-21, a related saponin that is currently the favored adjuvant in anticancer and antiviral vaccines. Tedious isolation and purification protocols have hindered the clinical development of QS-7. A novel semi-synthetic method is provided wherein a hydrolyzed prosapogenin mixture is used to synthesize QS-7, QS-21, and related analogs, greatly facilitating access to QS-7 and QS-21 analogs for preclinical and clinical evaluation.
