The disabling feature may be configured to be activated during operational motion of the plunger relative to the barrel, such as during insertion of or withdrawal away from a suppository. The device can include a structural element of the barrel or the plunger configured to engage with the disabling feature of the barrel or the plunger during insertion of or withdrawal away from the suppository to activate the disabling feature. The disabling feature may be implemented in a number of ways to ensure single-use operation of the single-use suppository insertion device.

Embodiments also provide various drug preparations that are configured to be contained within the capsule, advanced from the capsule into the intestinal wall and degrade to release the drug into the bloodstream to produce a therapeutic effect. The preparation can be operably coupled to delivery means having a first configuration where the preparation is contained in the capsule and a second configuration where the preparation is advanced out of the capsule into the intestinal wall. Today's suppositories are typically packaged individually for digital administration into the anal canal of a patient. A suppository applicator described herein, which works with individually packaged suppositories, can alternatively be shipped in a prepackaged operational arrangement with a suppository to enable the patient to administer the suppository through use of the applicator in a single operation after opening a hygienic wrapper containing the applicator-suppository combination. A method for manufacturing a suppository includes manufacturing the suppository in the presence of and in contact with an element configured to be used to insert the suppository into a body cavity. The method may include using the element to define a shape of the suppository during the manufacturing. The resulting applicator-suppository combination is more hygienic and easier to handle than the separate applicator and suppository and provides more efficiency from time of opening the packaging to time of commencing administration of the suppository.

The method includes localized delivery of a material which contains a therapeutically effective agent and/or moiety. The localized delivery of the agent is achieved using a catheter-based delivery system and an implant. An implant can include a support component, a cover component, and a valve component. The valve component can be operable to permit flow through the implant. The implant can be delivered on a catheter, with one of the ends of the implant being selectively expanded to permit temporary occlusion of a vessel while delivering a material through the valve component to a downstream target region. The other end of the implant can thereafter be released such that the implant occludes the vessel, or the implant can be removed from the vessel entirely.

In one embodiment, the method includes positioning one or more solid drug units upstream of the drug delivery device; and driving the drug units into the drug delivery device with a flow of pressurized gas. The systems and methods may be used to load a drug delivery device that includes a device body, a number of solid, compressed drug tablets, and a retention frame. The device body includes a drug reservoir lumen and a retention frame lumen. The number of solid, compressed drug tablets is positioned in the drug reservoir lumen, and the retention frame is positioned in the retention frame lumen.
