Using Alternating Electric Fields to Increase Permeability of the Blood Brain Barrier

Certain substances (e.g., large molecules) that ordinarily cannot traverse the blood brain barrier can be introduced into the brain by applying an alternating electric field to the brain for a period of time, wherein the frequency of the alternating electric field is selected so that application of the alternating electric field increases permeability of the blood brain barrier. In some embodiments, the frequency of the alternating electric field is less than 190 kHz (e.g., 100 kHz). Once the permeability of the blood brain barrier has been increased, the substance is able to cross the blood brain barrier.

A system for treatment of wounds consists of a treatment housing, a fluid delivery mechanism for supplying debridement fluids to the wound treatment area, an evacuation mechanism for evacuation of debris from the treatment chamber. A handheld device is connected to the treatment housing, wherein its interior accommodates a laser source, a scanning device, an image recording device and at least one sensor/detector, a control unit having a microprocessor for controlling operation of the system. The sensor detects concentration of various substances in the wound, and the microprocessor analyzes data obtained by the sensor and generates signals to adjust parameters of the laser, the liquid dispensing nozzles and the suction outlet to optimize removal of necrotic tissue so as to ultimately to promote wound healing.

SYSTEMS AND METHODS FOR OPENING TISSUES

The disclosed system can include a navigation guidance device configured to locate and/or monitor the target tissue, a single-element transducer for stimulating the target tissue with focused ultrasound (FUS), and a processor configured to determine a cavitation mode. The navigation guidance device can include a cavitation detector and an arm. The single-element transducer can be attached to the arm and be configured to induce the FUS with a predetermined parameter to open the target tissue.

Compositions, methods and systems for gas vesicle based cavitation

The system and process of therapeutic and effective cavitation by using ultrasound to collapse gas vesicles as well as cavitate the bubbles produced from the collapsed gas vesicles. Therapeutic effect includes, but is not limited to lysing cells by cavitation. The cells expressing the gas vesicles can optionally be used as delivery cells to preform tasks such as transporting the gas vesicles into deep tissue areas, releasing compounds at the cavitation site, and more. The gas vesicles can optionally be modified to facilitate getting the bubbles near the cavitation targets by functionalizing the gas vesicles.

Transfection and drug delivery

An ultrasound transmitter device for treating a patient is provided. The ultrasound transmitter device includes an imaging probe; an imaging array; and a therapeutic ultrasound device, wherein the imaging probe is configured to guide the therapeutic ultrasound device to the patients treatment site by use of ultrasound imaging with the imaging array, wherein the therapeutic ultrasound device is configured to produce a controlled intensity of ultrasound energy for treating the patients treatment site, and wherein the imaging probe and the therapeutic ultrasound device are configured to work in conjunction with one another to apply therapeutic ultrasound to tissue or bone graft sites in the patient.

MICROBUBBLES

The present invention provides methods for preparing acoustically-sensitive microbubbles. The method includes the steps of: i) preparing a first surfactant solution comprising a first micelle-forming surfactant at a concentration above the critical micelle concentration (CMC); ii) adding one or more pharmaceutical compounds in a solvent to the first surfactant solution, thereby loading the micelles with the one or more pharmaceutical compounds; iii) preparing a second surfactant solution comprising a second surfactant, wherein the second surfactant comprises one or more matrix forming surfactants; iv) adding heat to the second surfactant solution to melt the surfactant and allowing the mixture to cool under rapid stirring; v) combining the second surfactant solution with the loaded micelles; vi) purging the surfactant mixture with a purging gas; vii) agitating the purged mixture under a constant stream of the purging gas; and, viii) separating the formed microbubbles by size.

Intraarterial (IA) Application of Low Dose of Ethyl Alcohol Enables Blood-Brain Barrier (BBB) Impermeable Therapeutics Brain Entry
20220287986 · 2022-09-15 ·

A method of inducing blood-brain barrier (BBB) opening of for delivery of a non-BBB permeable substance to a subject includes the steps of: administering an ethyl alcohol at 0.015%-5% v/v into a blood stream of the subject via intra-arterial injection, intracardiac injection, or intra-arterial injection guided by ultrasound, wherein said ethyl alcohol reaches a brain of the subject to induce opening of blood-brain barrier so that the non-BBB permeable substance is capable of penetrating the blood-brain barrier to reach the brain of the subject. The non-BBB substance includes a therapeutic agent, a diagnostic agent, or a prophylactic agent. The use of low concentration of ethyl alcohol can induce a temporary BBB opening within 240 minutes after administration so that the non-BBB substance can be deliver to the brain during the time frame of BBB opening. Since ethyl alcohol is water soluble, the non-BBB substance can also be administered concurrently.
