COMPOSITION AND METHOD FOR BUCCAL ADMINISTRATION OF GnRH AGONISTS
20170348231 · 2017-12-07 · ·

Provided herein is an optimized method of enhancing endogenous gonadotropins and testosterone/estrogen production and concentrations comprising administration of a therapeutically effective amount of at least one GnRH agonist to a patient in need of such treatment. A method and dosing regimen is also described for delivery across the mucosa of the oral cavity for maximizing absorption and delivery, limiting systemic exposure and unwanted side effects, effectuating a sustained increase in concentrations of sex hormones and providing a non-invasive delivery that does not require inpatient visits. The method described benefits conditions such as hypogonadism in men due to aging or disease and symptoms related to menopause in women.

PHARMACEUTICAL COMPOSITIONS AND RELATED METHODS OF DELIVERY

The pharmaceutical compositions described herein include a suspension which comprises an admixture in solid form of a therapeutically effective amount of a therapeutic agent and at least one salt of a medium chain fatty acid and a hydrophobic medium, e.g. castor oil or glyceryl tricaprylate or a mixture thereof. The pharmaceutical compositions described herein contain medium chain fatty acid salts and are substantially free of alcohols. The pharmaceutical compositions may be encapsulated in a capsule. Methods of treating or preventing diseases by administering such compositions to affected subjects are also disclosed.

Implantable prosthetic device comprising a cell expressing a channelrhodopsin

The present invention provides compositions and methods for light-activated cation channel proteins and their uses within cell membranes and subcellular regions. The invention provides for proteins, nucleic acids, vectors and methods for genetically targeted expression of light-activated cation channels to specific cells or defined cell populations. In particular the invention provides millisecond-timescale temporal control of cation channels using moderate light intensities in cells, cell lines, transgenic animals, and humans. The invention provides for optically generating electrical spikes in nerve cells and other excitable cells useful for driving neuronal networks, drug screening, and therapy.

TREATMENT OF HORMONAL DISORDERS OF GROWTH

The present invention refers to a GPR101 inhibitor, antagonist or inverse agonist or inverse agonist for use in preventive and/or therapeutic treatment of diseases selected from the group consisting of acromegaly and gigantism and to methods for preventive and/or therapeutic treatment of diseases selected from the group consisting of acromegaly and gigantism wherein to a subject GPR101 inhibitor, antagonist or inverse agonist is administered. Further, the present invention provides a GPR101 agonist for use in preventive and/or therapeutic treatment of disorders selected from the group consisting of dwarfism, short stature, hypopituitarism and a disease of low levels of pituitary hormone secretion and to methods for preventive and/or therapeutic treatment of diseases selected from the group consisting of dwarfism, short stature, hypopituitarism and a disease of low levels of pituitary hormone secretion wherein to a subject GPR101 agonist is administered. The present invention also provides GHRH inhibitors, antagonists or inverse agonists and GH antagonists for use in the therapeutic treatment of X-linked acrogigantism (X-LAG syndrome). In addition, the present invention refers to a method of increasing body mass and/or body size of livestock comprising administering to livestock an effective amount of GPR101 agonist. Further, the present invention is directed to a non-human transgenic animal, comprising as expressed transgene a gene encoding GPR101 or overexpressing endogenous GPR101 gene.

PEPTIDOMIMETIC MACROCYCLES

The present invention provides biologically active crosslinked polypeptides with improved properties relative to their corresponding precursor polypeptides, having good cell penetration properties and reduced binding to human proteins. The invention additionally provides methods of identifying and making such improved polypeptides.
