The Method of Producing Sterile Cannabis sativa L. Cultivars includes applying a novel colchicine treatment to axillary meristems of diploid cannabis plants to produce sterile cannabis growth. The colchicine may be a range of concentrations applied over a 24 hour period to produce the sterile cannabis growth. The sterile cannabis plant will not reproduce sexually but may be replicated via other methods that are used to replicate plants.

The present invention relates to a method for producing sterile flowering biomass, the method comprising: a) inoculating a temporary liquid immersion culture system containing a sterile culture medium containing at least one cytokinin with sterile plant material, b) growing leafy biomass, and c) inducing and maturing flower tissues from the leafy biomass, wherein flowering is induced by reducing the duration and/or frequency of the immersion of the plant material in the culture medium in step c) compared to step b). It further relates to a method for producing a component present in flowering biomass, using sterile flowering biomass obtained using the aforementioned method according to the invention.

An economical and accurate method for identifying sex of a Cannabis plant involves amplifying an amplicon of genomic nucleic acid molecules from a subject Cannabis plant using polymerase chain reaction (PCR), the amplicon being a genomic region containing the nucleotide sequence as set forth in SEQ ID NO: 1. The sex of the subject Cannabis plant is determined by determining whether SEQ ID NO: 1 is SEQ ID NO: 2 or SEQ ID NO: 3, where SEQ ID NO: 2 is in male plants and SEQ ID NO: 1 is in female plants. Preferably, high resolution melt (HRM) analysis is used to determine the sex of the subject Cannabis plant by comparing the HRM analysis to a control for male or female Cannabis plants. The amplicon can be amplified in both male and female plants with a common primer set.
