Assay cartridges and methods of using the same

Assay cartridges are described that have purification, reaction, and detection zones and other fluidic components which can include sample chambers, waste chambers, conduits, vents, reagent chambers, reconstitution chambers and the like. The assay cartridges are used to conduct multiplexed nucleic acid measurements. Also described are kits including such cartridges, methods of using the same, and a reader configured to analyze an assay conducted using an assay cartridge.

CELL SORTING DEVICES

In one example in accordance with the present disclosure, a cell sorting device is described. The cell sorting device includes a microfluidic channel to serially transport individual cells from a volume of cells along a flow path. A sensor disposed in the microfluidic channel distinguishes between a cell to be analyzed and waste fluid. The cell sorting device includes at least two fluid transport devices disposed within the microfluidic channel. The at least two fluid transport devices include a cell ejector to, responsive to detection of a cell to be analyzed, eject the cell to be analyzed from the cell sorting device and a waste transport device to direct the waste fluid to a waste reservoir.

Fluidic system, use, and method for operating the same

A fluidic system having a first volume, a second volume and a membrane geometrically separating the two volumes, which has an open-pore microstructure for the passage of a first medium and a second medium. There is a contact angle (Θ) between the interface of the media and the pore surface. A first electrical field in the region of the membrane and a first electromagnetic radiation and a first heating of the membrane define a first state (Z.sub.1), in which the membrane is not wetted or is less wetted by the first medium and is more heavily wetted by the second medium such that a first contact angle Θ.sub.1>90° is formed between the pore surface and the interface. The first medium and the second medium and the pore surface have a surface energy of which at least one surface energy can be reversibly changed in such a way that a second contact angle Θ.sub.2<Θ.sub.1 occurs between the pore surface and the interface in a second state (Z.sub.2).

APPARATUS AND METHOD FOR CLAMPING A MICROFLUIDIC DEVICE

An apparatus suitable for clamping at least one microfluidic device, which includes (i) a fluid-tight chamber having a fluid inlet, the chamber being configured to receive a microfluidic device to be clamped by compression of at least one deformable part of the microfluidic device under the action of a pressure of a clamping fluid in the chamber, and (ii) a perfusion fluid management system configured to adjust the pressure of a perfusion fluid in the microfluidic device in such a way that, during a clamping operation, the pressure of the clamping fluid in the chamber is strictly higher than the pressure of the perfusion fluid in the microfluidic device.

MODULAR MICROFLUIDIC DEVICE

Described is a modular microfluidic device (MMD) for producing an analyte composition from a biological fluid sample, said device comprising: (a) a reagent module (RM) comprising a reagent reservoir containing a reagent and an eluent reservoir containing an eluent, said reagent and eluent reservoirs being coupled to one or more RM microchannels; and (b) a sample preparation module (SPM) comprising a SPM microchannel adapted to couple with the RM microchannel whereby fluid continuity between SPM and RM microchannels is produced on coupling, and: (i) a sample inlet for receiving said biological fluid sample; (ii) an outlet for delivering said analyte composition; (iii) a mixing chamber; (v) a metering chamber; (vi) an eluent chamber; (vii) a valve; (viii) a solid phase extraction element (SPE); and (ix) an aspirator in fluid communication with the sample inlet of the SPM for withdrawing an aliquot of said biological fluid when contained in a sample vessel, said aspirator being operably coupled to a pneumatic fluid level sensor.

Modular robotic systems for delivering fluid to microfluidic devices

Systems and methods for delivering fluid to a microfluidic device is provided. The system includes a multi-well plate having a plurality of wells and an inlet tube having a first end being in communication with the one or more wells of the multi-well plate and a second end being in communication with a microfluidic device. The first end of the inlet tube is moveable between the plurality of wells of the multi-well plate to deliver fluid to the microfluidic device from the plurality of wells of the multi-well plate.

A system for actuating the mechanical valve comprises an actuation member configured to move the mechanical valve from an open position to a closed position and an armature connected to the actuation member. The armature is configured to engage an electromagnet, wherein the electromagnet can be switched between an active state in which it electromagnetically holds the armature and an inactive state in which it does not electromagnetically hold the armature. First biasing means arc disposed between the actuation member and a bearing surface, wherein the first biasing means is configured to bias the actuation member into a first position in which it actuates a mechanical valve in a fluidic cartridge inserted into the reader.
